Abstract
This study aims to determine the pressure of methane gas produced by the seaweed Eucheuma cottonii and characterization of methane-producing bacteria learn in the process of making biogas from Eucheuma cottonii seaweed species. Laboratory scale research conducted in July-August 2010. The material used is seaweed Eucheuma cottonii 5 kg and 10 kg of sediment taken from the coastal waters Kalianda, Lampung province. Making biogas and methane gas pressure calculations performed at the Laboratory of Marine Biology Department of Marine Science Sriwijaya University and characterization of bacteria carried in the Great Hall of the Public Health Laboratory Palembang. Measurement of methane gas pressure using a manometer and characterization of bacteria through the three stages of colony morphology, cell morphology and physiology with test observations of biochemical reactions. The results showed that the seaweed Eucheuma cottonii can produce methane gas with a maximum pressure of 14.90 psi on day 24. From the results obtained by characterization of the bacteria bacillus-shaped bacteria that are gram-negative group. In tests of biochemical reactions that function to see the metabolic activity of bacteria showed positive results in the motility test, glucose, maltose, indole, TSIA (H2S), red methil, citrat simmon\u27s, lysine decar and shown negative results in tests of lactose, mannitol, sucrose, urea, voges proskauers and ornithine decar
