Differential and Cytochemical Analysis of Blood Cells in Rusty Blenny (Parablennius sanguinolentus, Pallas 1814)

Abstract

Rad opisuje hematološku analizu pune krvi babice balavice Parablennius sanguinolentus. Utvrđena je brojnost eritrocita, trombocita, limfocita, neutrofila, monocita i bazofila. Morfološka i kvantitativna analiza krvnih stanica provedena je bojanjem May-Grünwald Giemsom. Diferencijalnim citokemijskim tehnikama napravljena je citološka karakterizacija razlicitih tipova stanica. Glikokonjugati i polisaharidi dokazani su tehnikama alcian blue (AB) i periodic acid Schiff (PAS). Stanicni lipidi dokazani su tehnikom sudan black B (SBB). Enzimatsko-citokemijskim tehnikama dokazala se aktivnost alfa-naftil acetat esteraza (ANAE), alfa-naftil butirat esteraza (ANBE) te alkalne fosfataze (ALP). Brojnost i udio leukocita, eritrocita i morfologija stanica slicni su ostalim vrstama iz reda Perciformes. Pozitivnu reakciju na bojanje ABom imaju jezgre svih stanica. PASom se boje limfociti, monociti, neutrofili i trombociti. SBBom se boje limfociti, neutrofili i monociti. Aktivnost ANAE nije pokazao niti jedan tip stanice. Aktivnost ANBE pokazuju limfociti, neutrofili i monociti. APFB je pokazala aktivnost kod neutrofila i monocita.This thesis presents results attained from peripheral blood of the fish Parablennius sanguinolentus. It contains erythrocyte, thrombocyte, limphocyte, monocyte, neutrophile and basophile counts. The May-Grünwald Giemsa stain was used in morphological and quantitative blood cell analysis. Cytological characterization of all blood cell types used differential cytochemical stains. The alcian blue (AB) dye and the periodic acid Schiff method (PAS) stained glycoconjugates and polysacharides. The sudan black B (SBB) dye stained cell lipids. Activities of alpha-naphthyl acetate esterase (ANAE), alpha-naphthyl butyrate esterase (ANBE) and alkaline phosphatase (ALP) were proved using enzyme-cytochemical reactions. Leukocyte and erithrocyte count, percentage and cell morphology are similar to other fish species from order Perciformes. AB stained nuclei of all cells. PAS stained lymphocytes, monocytes, neutrophils and thrombocytes. SBB stained lymphocytes, neutrophils and monocytes. ANAE did not show reaction in any type of cell. ANBE stained lymphocytes, neutrophils and monocytes. APFB showed reactions in neutrophiles and monocytes
