[2+21-Cycloadducts of enamines and DMAD, formed in apolar solvents, isomerise to pyrrolizine derivatives.under mild conditions in protic polar sol- vents Zike methanol and I-butanot. Recent studies in our laboratories have revealed that the well known reaction of pyrrolidine enamines with electron-deficient acetylenes like dimethyl acetyle- nedicarboxylate (DMAD) only give cyclobutenes in solvents of low polarity. In po- lar solvents such as acetonitrile, nitromethane and particularly methanol we found that the same reactants give pyrrolizine derivatives'. The same difference was observed in reactions of 3-(1-pyrrolidinyl)thiophene derivatives with DMAD2. A qualitative comparison of the rates of these reactions in polar and apolar sol- vents learned that the rate varies with the electron density of the enamine dou- ble bond, but hardly with the polarity of the solvent in which the reaction is carried out3. This is strikingly different from the results obtained by Huisgen et aZ.5 for the reaction of an electron-deficient olefin (tetracyanoethylene) with electron-rich olefins (enol ethers). They found that the rate of the [2+23- cycloaddition reactions is strongly dependent on the polarity of the solvent (k (acetonitrile)'k(cyclohexane) 1-6~10~) and they have attributed this to the stabilization of the-transition state of the rate-determining step, which in- volves the formation of a 1,4-dipolar intermediate. Since the reaction of pyr- rolidine enamines and DMAD in polar solvents to give pyrrolizines proceeds most likely via a 1,4-dipole, the absence of a substantial solvent effect on the rate let us to suggest that the rate-determining step in the reactions of pyrrolidine enamines and DMAD might be the formation of a tied-ion pair. That different pro- ducts are obtained in polar and apolar solvents can be rationalized assuming that in polar solvents the tied-ion pair is rapidly converted into a pyrrolizine, via a 1,4-dipolar intermediate which is stabilized by solvation in polar solventsl. In this communication we describe the conversion of 3-(1-pyrrolidinyl)cyclobutene derivatives into pyrrolizines in polar solvents. 3-(1-Pyrrolidinyl)cyclobutenes generally are reactive compounds, since they 1217 1218 easily undergo ring opening often at temperatures below 20°6. However, compounds l-3 have been prepared by reaction of the corresponding enamines with DMAD in -- II CH3 N ““tpc: 1 CH2E E = COOCH3; E’= COCC(CH3)3 apolar solvents such as diethyl ether or benzene'-9. Under similar conditions the reaction of the pyrrolidine enamine of a-tetralone and di-tert-butyl acetylenedi- carboxylate in acetonitrile gave 4 lo in a yield of 83% [mp 104-105°; m/e 425.257 (M+); 1~ NMR G(CDC13) 3i40-3.60 (m,l~,-;H-1, NMR G(CDC13) 65.5 (s,N-C-1, 40.8 (a,-&H-)1. 1.51 and 1.38 (s,9H,C(CH3J3); 13C When the [2+21-adduct i7 was dissolved in methanol at 20° we observed a quan- titative conversion (2 95%) into the corresponding pyrrolizine 2 within two hours Coil; m/e 325.132(M+); lH NMR G(CDC13) 4.85 (dd,J=6 and ;OHz,lH,N-kH-1, 3.23 and 2.67,(AB-q,J=17Hz,2H,CH2E); 13C NMR G(CDC13) 155.2 (SIN-C=), 76.8 (d,N-AH-), 54.4 (s,-C(E)CH2E)I. The same pyrrolizine was obtained by reaction of 2,3-dihydro-3,3- dimethyl-4-(1-pyrrolidinyl)thiophene and DMAD in methanol at O" in a yield of 37%. In a similar way compound 2 was formed within two hours when 2 was dissolved in - methanol, in a yield of 78% Cmp 114-115O; m/e 327.147 (M+); lH NMR G(CDC13) 4.91 (dd,J=6 and 9Hz,lH,N-AH-) 157.5 (s,N-k=) , 3.24 and 2.78 (AB-q,J=17Hz,2H,CH2E); 13C NMR G(CDC13) , 77.9 (d,N-&H-j , 54.8 (s,-&E)CH~E)I~~. Isomerization of compound 1219 2 required heating for 24 hours in methanol at reflux temperature. We obtained a 3:5 mixture of the pyrrolizine 7 [mp 117.5-118.5°; m/e 425.255(M+);'HEMR S(CDC13) 4.75 (dd,J=6 and 9Hz,lH,N-;H-), G(CDC13) 146.9 (s,N-&=I, 3.35 and 2.51 (AB-q,J=17.5Hz,2H,CH2d; 13C NMR 70.5 (d,N-;H-,, 57.0 (s,<(~CH2E? ]I2 and the benzocyclo- octene 2 [mp 131.5-133.5O; m/e 425.253(M+); NMR G(CDC13) 157.2 (s,N-;=I, lH NMR G(CDC13) 6.28 (m,lH,=CH-1; 13C 136.8 (d,-CH=)l. Pyrrolizine 1 could also be prepar- ed by reaction of the enamine and the acetylene in methanol at 65O for 16 hours in a yield of 41% and 2 was obtained almost quantitatively by heating 2 in toluene for 2.5 hours at llO". We can explain these isomerization reactions of the cyclobutene derivatives to pyrrolizine derivatives in polar solvents in terms of a conversion of the cyclo- butene in a strongly solvated 1,4-dipolar intermediate 11 (Scheme)13. Subsequent transfer of two hydrogen atoms gives the 1,5-dipole 12 which cyclizes to give a - pyrrolizinel. Scheme In the literature8pgs15 isomerization reactions of [2+21-cycloadducts of ena- mines and acetylenic esters to the corresponding Michael adducts (structures of type 10) have been reported. Brannock et az. - g have explained this reaction by a [2+21-cycloreversion followed by a slow, irreversible formation of the Michael adduct. We have repeated such a reaction viz. the isomerization of methyl l-(l- pyrrolidinyl)bicycloC3.2.03hept-6-ene-7-carboxylate in methanol and observed af- ter 45 hours at 20° a 1:5 mixture of the starting material and the Michael ad- ducts (E:Z=2:1). In view of theseresults, the Michael adduct obtained in methanol by Eberbach and CarrE4 might well have been formed via the [2+21-cycloadduct, since the rates of formation are the same. During the isomerization of 1 to 2 the lH NMR spectrum showed a transient sin- glet absorption at 6 5.95. When the reaction of the pyrrolidine enamine of a-tet- ralone and DMAD in methanol at O" was stopped after 15 minutes the lH NMR spec- trum of the reaction mixture exhibited two signals at 6 6.33 and 6 5.95 respec- tively but after a prolonged reaction time these signals disappeared and we only isolated the known pyrrolizine gl. We assign the signals at 6 6.33 and 6 5.95 to the E- and Z-isomers of the Michael adducts 10 respectively, because these val- - ues are in agreement with values reported for such compounds16. Therefore it seems that in the isomerization of 3 to - 2 the Michael adduct is an intermediate. 1220 Acknowledgement. We have measured the rates of the reaction of dimethyl 4-(1-pyrrolidinyl)-2,3- thiophenedicarboxylate with DMAD at 100° both in 1-butanol and in toluene and we found that the reaction in the polar solvent is only about twenty times faster than that in the apolar solvent. Eberbach and Carre4 also reported no difference in the rate of reaction of a dihydroazepine with DMAD in solvents of different polarity although in acetonitrile the rearranged [2+21-cycload- duct is formed exclusively, whereas in methanol only the Michael adduct was obtained. Satisfactory elemental analyses were obtained for all new compounds (C,H,N 2 0.3%). Compound 5 was formed independently in the reaction of the pyrrolidine ena- mine of a-indanone and DMAD in methanol at O-5'. The NMR spectra of the pyrrolizines 1 and 21 are similar. Huisgen et al.14 have intercepted a 1,4-dipole when the acetone solution of the cyclobutane, derived from ethyl vinyl ether and tetracyanoethylene was kept for one week at room temperature. R.
