主动靶向型小干扰核糖核酸输送载体及其制备方法

Abstract
本发明公开了一种主动靶向型小干扰核糖核酸输送载体,由作为载体骨架的可生物降解的高分子化合物分别与作为载体侧链的靶分子和氨基化合物共价偶联得到。该载体能与siRNA自组装形成复合物纳米粒,将siRNA紧密包裹起来,防止核酸酶对siRNA的降解。另外,该载体中的靶分子能与肿瘤细胞表面的抗原或受体发生特异性相互作用,从而实现将载体-siRNA复合物纳米粒精准地靶向输送至肿瘤部位,通过抗原或受体介导的细胞内吞作用进入癌细胞,在胞内通过siRNA诱导基因沉默复合物的生成,序列特异性降解mRNA,使目的蛋白表达量下调,从而达到杀死癌细胞,实现治疗肿瘤的目的。本发明还提供了该主动靶向型小干扰核糖核酸输送载体的制备方法
