paper, thus removing the need for x­ray film and darkrooms. The kit is relatively cheap,safe and easy to use. Digoxigenin­labelled probes are stable enough to be usedrepeatedly for a year or longer and can be preserved in ethanol and sent by ordinary mailto laboratories. This digoxigenin labelling of DNA will simplify and increase the utility ofparasite identification based on recombinant DNA probes.Identifying lethal components of the Theileria disease complexTO IMPROVE CONTROL of tick­borne diseases, scientists need a clear understandingof how they are transmitted in the field. In the case of East Coast fever, for example, theyneed to know where, when and to what extent Theileria parasites occur in tick, wildanimal and cattle populations.Complicating such epidemiological studies are the existence of several species ofTheileria, not all of which cause disease. Theileria taurotragi, for example, is a benignspecies that is indistinguishable in structure in the tick vector from Theileria parva, aparasite that causes lethal infections in cattle. The problem of distinguishing non­pathogenic Theileria parasites from mildly and severely pathogenic organisms has, untilrecently, been a stumbling block to epidemiological research.USE OF MOLECULAR BIOLOGY techniques is solving that problem. ILRAD scientistshave cloned DNA sequences of Theileria parasites for use as 'molecular probes' toidentify genomic differences among Theileria species. They have now developedsecondary probes using synthetic oligonucleotides based on identified parasite DNAsequences. Probes made of repetitive parasite DNA sequences were produced atILRAD several years ago to distinguish T. parva and T. mutans from other Theileriaparasites.More recently, in collaboration with scientists supported by the British OverseasDevelopment Administration and scientists at the Kenya Agricultural Research Institute(KARI), ILRAD staff developed a panel of probes targetted to parasite ribosomal RNAsequences. Use of these probes is enabling scientists to distinguish T. parva species withunequalled precision, including two species economically important in Africa and Asia, T.parva and T. annulata, respectively, T. mutans, which is mildly pathogenic, and thebenign T. taurotragi.USING THE NEW PROBES, scientists from ILRAD and Japan's Tropical AgriculturalResearch Centre (Tsukuba) recently determined that a species of Theileria isolated byKARI scientists and causing disease in cattle in Kenya (Naivasha) is Theileria buffeli, aparasite formerly known to occur only in Asia, where it also causes disease. It is not yetknown what species of tick transmits this parasite in Africa, but the discovery of itsoccurrence on the continent will help researchers to establish the dynamics of tick­bornediseases and to predicate more accurately where the diseases may spread.Role of macrophages in development of anaemia intrypanosomiasis (Ph.D.Thesis)The engulfing of erythrocytes (red blood cells) by macrophages is known to be one of theprimary causes of anaemia in animal trypanosomiasis. To analyse the mechanisms of thephagocytosis of erythrocytes, an in vitro assay was established in which peripheral bloodmonocytes were purified and incubated with 51Cr­labelled erythrocytes. The release of51Cr was used as a measure of erythrophagocytosis.Macrophages from trypanosome­infected cattle had a greater rate of erythrocytedestruction than those from uninfected cattle. Erythrocytes from infected cattle were moreprone to destruction by macrophages from both trypanosome­infected and uninfectedcattle. Erythrophagocytosis by peripheral blood­derived macrophages, bone marrowmacrophages and spleen macrophages was examined at eight time points inTrypanosoma congolense­infected Boran calves and found to increase from 14 dayspost­infection, reaching a maximum by 35 days post­infection. In vitroerythrophagocytosis was greatest in splenic macrophages.In vitro macrophage erythrophagocytosis increased following T. congolense infection insix Boran cattle, six N'Dama cattle and one buffalo. The rates were significantly higherand occurred earlier in the Boran cattle than in the N'Dama; levels in the buffalo were thelowest. There was a more rapid release of Cr51 by macrophages of Boran cattle than ofN'Dama cattle, which suggests that Boran macrophages engulf more erythrocytes overtime than N'Dama. The Boran cattle became more severely anaemic than the N'Damacattle. Three Boran and three N'Dama cattle infected with T. vivax had increasing butsimilar rates of erythrophagocytosis throughout infection. This correlated with levels ofanaemia in the two groups, which dropped at similar rates.To investigate the possible mechanism(s) of macrophage activation anderythrophagocytosis during bovine trypanosome infections, macrophages were exposedto trypanosome antigens and mammalian immunomodulatory factors and the levels oferythrophagocytosis were measured. Macrophages incubated with trypanosome lysatebut not bacterial li
