Due to theirspecial physicochemical properties, fullerene-based amphiphilescan be used to create nanoscale supramolecular structures.2 Con-jugation of hydrophilic entities onto the surface of hydrophobicfullerenes is one way to prepare fullerene based amphiphiles.3,4Alternatively, they can be synthesized by attaching fullerene salts tohydrophobic segments.5 One of the most relevant properties of theamphiphilic materials is that the fullerene-based amphiphiles canself-assemble into diﬀerent types of nanostructures, depending onsuch physicochemical factors6 as concentration, polarity of themedium, and the chemical structure of the molecule conjugatedto the surface of the fullerene.7 The last factor not only aﬀects thestructure of self-assemblies of fullerene-based amphiphiles andtheir physiochemical properties, but also dramatically dominatestheir biological behavior. For example, conjugation of fluorouschains onto the surface of fullerenes results in the formation ofspherical vesicles in which nonpolar fluorous chains are exposed toan aqueous environment,8 while dodeca-dendronized fullerenesself-assemble into a disc-shaped structure where the fullerenes arelocated in the center.9 Hydrophilic macromolecules conjugated tofullerenes, on the other hand, improve their biological propertiesfor biomedical applications.10,11 Furthermore, self-assembliesof fullerene-based amphiphiles can be fixed in the form ofstable nanostructures by irradiation with UV light.12 Thisstrategy is a promising approach to produce nanocarriers withcontrolled permeability and transport properties.However, the critical issues for obtaining stable fullerene-basednanostructures with extraordinary transport properties are the typeof hydrophilic polymer and a straightforward method to conjugatethis polymer to the surface of fullerene. Photocrosslinking theassemblies of fullerene-based amphiphiles should not aﬀect theother blocks, just the fullerene segments. Therefore, polymerswithout any significant absorption in the UV region are suitablecandidates in this case.Polyglycerol has been used as a highly water soluble, multi-functional, and biocompatible compound to prepare a widerange of nanostructures for promising nonmedical tools.13–15Conjugation of polyglycerol to carbon-based nanomaterials notonly enhances their water solubility and multi-functionality butalso improves their biocompatibility.16,17 Moreover, it is almosttransparent in the UV-vis region, which allows photocrosslinkingof the fullerene-polyglycerol assemblies and production of nano-capsules in the aqueous solutions. Based on these envisionedoutcomes, polyglycerol is a suitable candidate for producingfullerene-based amphiphiles that are able to create nanostructureswith the new transport properties through molecular self-assemblyand photocrosslinking in the aqueous solutions.In this work, FPAs were synthesized on the gram scale usinga one-pot ‘‘grafting from’’ method. The easy production ofsuch amphiphiles is a big advantage for their practical applica-tion. Photocrosslinking the oligomeric assemblies of FPAs inaqueous solution resulted in nanoparticles with unusual trans-port properties. In addition, confocal microscopy showed thatphotocrosslinked assemblies are able to hold encapsulateddyes in a physiological medium for a long time withoutsignificant leakage.Nucleophilic reaction between potassium tert-butoxide andfullerene led to tert-butoxy functionalized fullerenes with anioniccenters that were able to initiate the anionic ring-openingpolymerization of glycidol (Scheme 1).a Institut fu¨r Chemie und Biochemie Organische Chemie, Freie Universita¨t Berlin,Takustr. 3, 14195 Berlin, Germany. Commun., 2016, 52, 4373--4376 This journal is©The Royal Society of Chemistry 2016The yield of reactions was high and in the range of 80–90%.For example, the mass of the purified product of the reactionthat was started using 4.4 g of glycidol and 0.3 g of fullerene was4 g, which confirms the gram-scale production of the FPAsusing this approach.These new FPAs were fully characterized using diﬀerentspectroscopic and microscopic methods (see ESI†). The numberand molecular weight of polyglycerol branches, conjugated to thesurface of fullerene, depended on the potassium tert-butoxide/fullerene and glycidol/fullerene ratios, respectively (see Table S1and Fig. S3, ESI†). The number of polyglycerol branches attachedto the fullerene segment was calculated using tert-butoxide/polyglycerol peak area ratios in the 1H NMR spectra (Table S2,ESI†). Keeping the glycidol/fullerene ratio constant, increase inthe potassium tert-butoxide/fullerene ratio resulted in a highernumber of polyglycerol branches attached to fullerene. Whenthese parameters were fixed in a ratio of 0.08 mol/0.4 mmol and0.8 mmol/0.4 mmol, respectively, the produced FPAs were easilysoluble in PBS, but had a high tendency to form aggregates. Thecritical aggregation concentration (CAC) of FPAs was measuredat 14 mM using the DLS method.18 Above CAC (120 mM), theyformed oligomeric assemblies with a hydrodynamic diameter of20 nm (Fig. 1 and Fig. S7a, ESI†).In order to verify the morphology and size of the oligomericassemblies, we performed cryo-TEM of the FPAs in PBS abovethe CAC. Fig. S8a (ESI†) shows a monodisperse distribution ofspherical particles with a mean diameter of 6 nm thought tocorrespond to the oligomeric assemblies (3–4) of FPAs.Transport capacity (TC) and transport eﬃciency (TE) offullerene-polyglycerol oligomeric assemblies for hydrophobicdyes, such as Cy5-CH3, were much higher than those for thehydrophilic dyes, such as Cy5-COOH (Table S3, ESI†).Based on cryo-TEM images, FPAs were thought to formoligomeric micelle-like assemblies, probably via hydrophobicinteractions of the unfunctionalized fullerene domains, wherebya central hydrophobic core could encapsulate hydrophobicguest molecules (Fig. 2a). Therefore, they were able to transportCy5-CH3 (Fig. 2b), whereas little transport eﬀect was observed forCy5-COOH (Fig. 2b).However, many therapeutic agents are hydrophilic compoundsand should be encapsulated and transported by the appropriatenanocarriers in order to improve their eﬃciency.19 Due to thedynamic nature of carriers that are assembled via weak noncovalentinteractions, they do not provide suﬃcient stability underphysiological conditions. Therefore, premature drug release andnonspecific biodistribution diminish their eﬃciency. Partial cross-linking of those nanocarriers, however, increases their stability andresults in improved transport capacities and also controls the releaseof the encapsulated therapeutic agents.20,21It has been proven that photocrosslinking of fullerenenanoparticles diminishes their permeability.12 Therefore, FPAs(120 mM) in PBS were irradiated by a UV USHIO super highmercury lamp (USH 102d, 100 W) to obtain partially crosslinkedfullerene-polyglycerol assemblies (PCAs) with improved encapsula-tion properties.The intensity of the lmax, which is characteristic for FPAs in theUV spectra and peaked at 280 nm, was almost reduced by half after3 h of irradiation in PBS, which evidences at least partial photo-crosslinking of the fullerene moieties to form aggregates (Fig. 3).Longer UV irradiation furthered the crosslinking process but didnot change the UV absorption of the maximum crosslinked (FCAs).Since the brown color of the neat FPAs solution is due to thep-system of fullerenes, the conversion of the color to yellow andlight yellow over the irradiation time made the gradual cross-linking of the fullerene segments visibly apparent (Fig. 3b and c).Scheme 1 Ring-opening polymerization of glycidol with a fullerene deri-vative that had anionic centers on its surface resulted in the formation ofFPAs. Commun., 2016, 52, 4373--4376 | 4375Cryo-TEM images show that photocrosslinking the oligomericassembly predominantly generated larger spherical assemblieswith diameters in the range from 100 up to 500 nm (Fig. 4a, band Fig. S8b, c, ESI†). The morphology of assemblies above 100 nmcan deviate from the spherical geometry in the images due to aflattening within the embedding vitrified solvent layer, which wasin the 100–200 nm range. Moreover, the magnified inset fromFig. 4b shows a stereo image (81-tilt) to prove the photolinkageeffect of FCAs (here in a linear fashion). Photocrosslinking of theaggregates was also evaluated by DLS measurements (see the ESI†for more details).Interestingly, crosslinking of the fullerene-polyglycerolassemblies by UV irradiation improved their encapsulationproperties towards Cy5-COOH, but partially diminished theirTC and TE for Cy5-CH3 (Fig. 5 and Fig. S9, S10, Table S3, ESI†).Obviously, after covalent crosslinking of the fullerene segmentsupon a [2+2] cycloaddition reaction, the hydrophobic part ofthe assemblies was less accessible to the hydrophobic dye.Therefore, the TC and TE for Cy5-CH3 changed inversely versusthe photocrosslinking time (Fig. 2c). Decreased permeability ofFCAs upon irradiation reduced diﬀusion or release of thehydrophilic dye from the hydrophilic environment and there-fore improved their TC and TE for Cy5-COOH (Fig. 2d).However, hydrophobic and hydrophilic dyes were stronglytrapped by FCAs and dye leakage from the nanoparticles in thephysiological medium was much lower compared to their non-crosslinked analogs, as was proven by in vitro release experimentsand cellular uptake study of Cy5-CH3 encapsulated by non-crosslinked assemblies (NCAs) and FCAs, respectively (Fig. S11,ESI†). These carriers were loaded with almost the same amountof dye (2  104 mM) and they were incubated with A549 cellswith concentration (10 mM) lower than the CAC of FPAs. SinceNCAs were not stable under these conditions, Cy5-CH3 did notencapsulate and showed a similar uptake to the free dye(Fig. 6b and c). Due to the covalent bonds, however, FPAs werestable even below the CAC and they were able to entrap the dye.Since they were larger clusters, as proven by cryo-TEM and DLS,their cellular uptake was supposed to be slow. Therefore, theuptake of encapsulated dye was likewise slow. Fig. 6d shows thelow fluorescence intensity of the encapsulated dye in compar-ison to the free dye or NCAs. Furthermore, cellular uptakestudies after 24 h incubation time did not show any significantleakage of the dye encapsulated by FPAs, which confirmed theFig. 3 (a) UV spectra of a 2 mg ml1 PBS solution of FPAs irradiated atdiﬀerent times by a UV lamp at 100 W. A549 cells were incubated with FPAs and NCAs loadedwith the same dye for 24 h. Lower fluorescence intensity in thecase of FPAs confirmed the low permeability of this nanocarrierand the slow release of the encapsulated dye (Fig. S15, ESI†).In summary, fullerenes can be used as a macroinitiator foranionic polymerization of glycidol, which leads to fullerene-polyglycerol amphiphiles. Size and transport properties ofthose amphiphiles in aqueous solution can be controlled viaphotocrosslinking of their hydrophobic fullerene segments.The SFB 765 collaborative research center and the corefacility BioSupraMol are gratefully acknowledged for financialsupport.
