Research on expression of HIV-1 env in baculovirus

Abstract

昆虫细胞杆状病毒表达系统(BEVS)是真核表达系统的一种,以一种无需额外辅助的重组昆虫杆状病毒作为表达载体,目前已被应用来表达不同来源的基因,如细菌、病毒、植物和哺乳动物的基因。它具有表达产量高,蛋白经过完善的后处理过程,如内含子剪切、糖基化等特点。其原理是通过外源基因置换病毒的核多角体蛋白基因,使外源基因在核多角体蛋白启动子下高效表达。本论文中采用BEVS表达HIV-1包膜蛋白,目的在于弥补原核表达系统不能表达大片段的、高度糖基化的重组蛋白;并探索其应用在检测试剂中的前景。Baculovirus / insect cell expression system (BEVS) is a kind of Eukaryotic expression system.
