Structural Studies of Nuclear Receptor Nur77 Dimerization

Abstract

核受体超家族是一类进化上相关并且能够与特定的DNA片段相互结合的转录因子。隶属于这一超家族的孤儿核受体Nur77,在各种不同刺激的诱导下,调控细胞生长、分化、凋亡,并在生物体发育的过程中发挥重要的作用,近年来已经成为一个重要的潜在药物靶点被广泛地研究。典型的核受体在受到特定的刺激下,若与相应配体结合,会以其配体结合结构域(LBD)为主发生同源或异源二聚化,并通过与DNA上被称之为应答元件的特定序列结合,发挥转录激活的生理功能。Nur77能够以单体、同源二聚体、异源二聚体三种方式分别与相应的应答元件结合,然而其具体的二聚化机理尚未被揭示。 第一个部分的研究中,我们研究已经被本实验室解析、具有高...Nuclear receptor superfamily consists of transcription factors that are evolutionarily related and can interact with specific DNA fragments.
