Study of Pelo deficiency mediated antiviral activity

Abstract

果蝇是研究抗病毒天然免疫以及病毒与宿主相互作用的理想模型。我们以果蝇C病毒(DrosophilaCVirus,DCV)为病原体对已有的突变体果蝇库进行筛选,找到了一个对DCV感染更为抵抗的果蝇株并鉴定出其体内Pelo基因的表达量下降。通过基因敲除和表型回复实验,我们证实了Pelo基因的缺失的确可以使果蝇对DCV的感染更为抵抗。此外,DCV以及其他多种病毒的复制在Pelo基因敲除型果蝇中都变慢,说明Pelo基因缺陷可能介导了广谱的抗病毒免疫。 我们研究发现,Pelo并不参与目前已知的果蝇抗病毒免疫途径,而是在蛋白质翻译水平特异性地调控DCVORF2蛋白的表达,并且这种调控不发生在蛋白翻译的起始...Drosophila is an ideal model for investigating antiviral innate immunity and virus-host interactions.
