Protein Kinase D3 Is Essential for Prostratin-stimulated Integrated HIV-1 Transcription activation via NF- κB Signaling Pathway

Abstract

Prostratin,一种非肿瘤促进性的佛波酯,能够通过激活HIV-1病毒转录来清除潜伏期的HIV-1病毒,但其分子机制还不是很清楚。本文,我们发现PKD3对于Prostratin激活HIV-1病毒转录是必需的。 首先,敲除PKD3能够阻断Prostratin对HIV-1病毒的转录激活,而敲除PKD家族的其他成员则不能。而且过表达持续活性的PKD3,而非野生型或没有催化活性的PKD3,能增强HIV-1病毒转录水平。于此同时,Prostratin通过增强PKD3活性结构域Ser731和Ser735位点的磷酸化促进PKD3的活性。另外我们鉴定出PKC𝜀是磷酸化PKD3的激酶。最后...Prostratin, a nontumorigenic phorbol ester, has been shown to be capable of eradicating the latent HIV-1 provirus by inducing HIV-1 transcription activation.
