The primary study of Hepatitis B virus core protein virus-like particle used as gene transfer

Abstract

基因治疗越来越成为一种治疗人类遗传病和癌症的一种手段,乙肝病毒核心蛋白病毒样颗粒具有规则的正二十面体结构,为基因治疗提供了新的载体选择。 本论文工作主要分为三个方面: 1、在构建好的乙肝病毒核心蛋白的大肠杆菌表达载体基础上,优化纯化步骤,探讨乙肝病毒核心蛋白病毒样颗粒(HBcVLPs)在体外条件下的解聚和重组条件,选定重组液的最佳配方,并考察解聚重组对HBcVLPs的形貌、抗原性等影响。 2、通过琼脂糖电泳等方法考察两种HBc-183和HBc-144这两种HBcVLPs对坍塌的质粒和未坍塌的质粒保护情况并通过荧光定量法测量VLPs对质粒的包封率。 3、通过转染报告基因pGL3,用化学发...Gene therapy has become more and more access into a cure of human inherited diseases and cancer.
