These phenomenahave attracted broad interest among a large community ofscientists that cross disciplines from mathematics, physics,chemistry, and biology to clinical scientists. DNA is con-tinuously exposed to a range of damaging agents, includingreactive cellular metabolites, environmental chemicals, ion-izing radiation, and UV light. The biochemical consequencesof DNA lesions are diverse and range from obstruction offundamental cellular pathways like transcription and repli-cation to fixation of mutations. Cellular misfunctioning, celldeath, aging, and cancer are the phenotypical consequencesof DNA damages accumulation in the genome. Fortunately,an intricate set of genome surveillance mechanisms functionto counteract genomic insults. Among these mechanisms,base excision repair and nucleotide excision repair areboth dedicated to the removal of single-strand lesionscontrary to double-strand break repair. Additionally, somespecialized polymerases can temporarily take over lesion-arrested DNA polymerases during S-phase, in a mutagenicmechanism called translesion synthesis. Such polymerasesonly work if a more reliable system, such as homologousrecombination, cannot avoid stumbled DNA replication.These DNA repair mechanisms function in conjunction withan intricate machinery of damage sensors, responsible ofa series of phosphorylations and chromatin modificationsthat signal to the rest of the cell the presence of lesionson the DNA. Together DNA repair mechanisms and DNAdamage signaling systems form a molecular shield againstgenomic instability called DNA Damage Response system.Hence, we have tried to integrate several papers that presenta synergy that emerge when researchers from diﬀerent fieldsput their forces together into a common goal, trying toimprove human health.
