The objective of this study wasto screen the differentially expressed genes in HIV/HCVco-infected patients, and further investigate its possiblemechanism.MethodsEligibility Criteria including 20-40 year-old male, similarsocial-economic background, treatment naive, sametransmission route methods (Sex or Injection drug), andCD4 count >200 cells/mm3. All research objects signedon informed consent. The project proceeded afterapproval of the local REC.Human blood samples were collected from 13 HIV/HCV coinfected patients, 8 mono infected patients, and8 normal control. At first, CD4 and CD8 positive T cellswere separated with magnetic activated cell sorting, thenfollowed by RNA extraction. After reverse transcriptedto cDNA with anchor primer and amplified with ran-dom primers, the products were run on PAGE electro-phoresis followed by silver stain. The bands withdifferent expression level were extracted from gel andcloned for sequencing. The expression changes wereconfirmed by SYBR Green real-time quantitative PCR.ResultsTotally 307 differentially expressed bands wereextracted. 25 fragments were sequenced and found 3 forring-finger protein 141(RNF141), 5 for cyclic AMPphosphoprotein (ARPP-19), 5 for mitochondrial cyto-chrome (cytochrome c and b-245). Most of the otherfragments were function unkown sequences located onX chromosome and mitochondrial. The change ofexpression of cytochrome c and ARPP-19 were con-firmed with a significant down-regulation in HIV/HCVcoinfected patients and a slight down-regulation inmono-infected group compared to negative population.Discussion1. The expression of RNF-141, ARPP-19, cytochrome cand b-245 mRNA are probably associated with HIV/HCV co-infection.2. The technique of mRNA differential displaytogether with SYBR Green quantitative real-time PCR isuseful for screening for differentially expressed genes inHIV/HCV co-infection.3. Since mitochondrial cytochrome has been reportedto associated with prognosis of AIDS. Is it possible toact as a useful marker for prognosis of HIV/HCV coin-fection?
