External tufted (ET) cells constitute onemajor class of excitatory JG interneurons, and recentexperimental studies indicate that they may play a centralrole in coordinating intraglomerular activity [1]. Amongtheir suggestive properties, ET cells have been found toburst endogenously at theta frequency, to entrain to pat-terned input at sniffing frequencies, and to synchronize viasynaptic and gap junctional mechanisms [2,3]. They alsoappear to be primary mediators of signal transmissionbetween olfactory receptor neurons and mitral cells [4].We present a single-compartment, Hodgkin-Huxley-style model of the ET cell which includes those identifiedmembrane currents which are known to be essential forendogenous bursting: spiking sodium and potassium cur-rents, a low-voltage activated T-type calcium current(ICaT), and a persistent sodium current (INaP) [5].A model variant incorporating an H-type current was alsoconstructed. Passive membrane properties were based ondata from the literature, while the kinetics of active cur-rents were obtained from voltage clamp data using the‘full-trace’ method of the NEUROFIT software package[6]. The model neuron (versions implemented in PyD-STool [7] and MATLAB) is designed for use in largerscale computational models of the juxtaglomerularcircuitry.We explore the model’s parameter space using detailedparameter sweeps and Latin hypercube sampling, demar-cating regions of bursting, tonic, and excitable behavior.To study the sensitivity of various functional characteris-tics to changes in parameter values, we employ specializedsmooth optimization methods for bursting neural models[8]. We find that the half-activation voltages for ICaT andINaP are most critical for control of interburst period andduty cycle. We also investigate the phase response proper-ties of the ET cell model and its responses to periodicinput.AcknowledgementsWe thank Michael T.
