In the point of view of commercial application of theartificial seed technology, the improvement of the dry type artificial seed tecbnologyseems to be one of the most promised approach (Redenbaugh, 1993). ln order todevelop e simple, and reliable protocol for laboratory-scale use, a new protocol ofdesiccation have been investigated based on PVC-foil covering method.Kanamyein-resistant (KmR) lines were obtained from cut somatic embryos of carrot(Daucus carota L. cv. Nantes Duke) by means of Agrobacterium tumefacienstransformatíon method using pGS gíuc-I cointegrate vector, following the protocolof Fári et alo (1994). Filter-uníformed, 500llm length early-cotyledon stage somatícembryos we.!eproduced from a line of KmR Nantes Duke according to the protocol ofSorvari et aI. (1994) without kanamicyn selection. Sodium-alginate-encapsulatedartificial seeds were produced following tbe method of Redenbaugh et al, (1986).Using an appropriate protocol of desiccation based on PVC-foil covering method(Fári et al., 1989), dry type artificial seeds were produced. Twenty artificial seedswere pIaced into each Petri-dishes of 10 em diameter and sea1ed with 50lJ.Illthin,commercial type PVC-foil (FOLP ACK, Hungary). The cultures have been exposed to30% external relative humidity (RH) in darkness at 25°C. The desiccation wasdetermined by weighting the water loss in three-hour-intervals, Afier 48 hours ofdesiccation, the process of rehydratation and germinated were carried ou! ou MS(1962) medium supplemented with 100 mgIliter kanamycin in Petri-dishes.According to OU! observation, the PVC-foil desiccation method of artificial seeds is asimple, and highly reproducible new method. Under the experimental condítíonused, the pattem of desiocation was linear. The speed of water loss have been veryslow, 4 mg/ h-I and afier 48 hours, ao average 15% water content was measured. Inthe experiment of germination, an average 60% of the dry type artificial seedsshowed good germination and their root-tips penetrated to lhe selection médiumcontaining kanamycin and they elongated intensely. To get homogeneousexperimental response, according to our data, the calibration of the artificial seeds ishighly important prior the desiccation phase.
