新疆哈密瓜转保鲜基因研究

Abstract
哈密瓜是新疆最具特色的瓜果之一,也是新疆最有影响的瓜果品牌。本研究试图通过转基因生物技术加速哈密瓜保鲜品种的培育,采用细菌来源的氨基环丙烷羟分子ACC脱氨酶基因导入哈密瓜直接降低乙烯合成前体ACC,抑制果实内乙烯合成,延缓哈密瓜采后的成熟时间达到保鲜的目的。成果在大量试验研究的基础上突破了相关的技术难题,建立了哈密瓜转保鲜基因的技术体系,获得了抗病毒和保鲜基因转化植株268株,转基因植株的移栽成活率达90﹪以上,获得果实2个。通过对21株基因转化植株的生理生化指标测定,发现伤后乙烯高峰期释放的乙烯比对照组降低80﹪以上,与其他研究结果比较间接提供了转ACC脱氨酶基因提高了哈密瓜保鲜期的效果
