Parentoni3 & EdilsonPaiva3The aim of this study is to obtain RFLP markers (genomic and cDNA) thatdiscriminate Al-tolerant and AI-susceptible maize inbred lines in order to mapgenes affecting this character and provi de better understanding about themechanisms of tolerance. The sources of DNA were two inbred lines L53 andLl327, previously identified as being tolerant and susceptible respectively. AIso,their FI hybrid was used. These genetic materiaIs have been developed by themaize breeding program of the National Maize and Sorghum Research Center -CNPMS/EMBRAP A. Ninety probes were selected at an average interval of 30cM in such a way to cover all the maize genome. These probes were then testedfor their ability to identify RFLPs between the parental inbred lines (L53 andLl327) when DNA was digested with Eco RI, Bam HI and Hind III. For ourhybridization work, a nonradioactive labeling system, using digoxigenin-dUTPand alkaline phosphatase proved to be quite efficient and reliable, resulting inSouthern blots with good resolution, alIowing the membranes to be stripped andreprobed at least three times. Thirty five probes showed codominant effectidentifying 55 RFLP loei that could distinguish the parental lines.
