This thesis is composed of three chapters: chapter I; general introduction, chapter II; study of PCI mechanism and chapter III; application of PCDR method to the study of cell differentiation mechanism in mouse embryos.  In general introduction, firstly, it is introduced about how studies on the use of light in therapeutic medicine have developed. Secondly, the importance of photosensitizers and its properties in photodynamic therapy (PDT) and PCI are discussed. Thirdly, PCI mechanism of cell penetrating peptide (CPP) – photosensitizer (PS) cargo conjugates are described and, later, application of PCDR and light dependent RNA delivery approaches are briefly reviewed.    In chapter II, after describing an introduction to the study of PCI mechanism, relation of PCI to heat, pH and Ca2+ ions are discussed. In this studied, the influence of heat in endosomal escape of CPP-PS/shRNA were investigated by changing the temperature of the TatU1A-Alexa546/shRNA treated cell. The correlation of PCI and endosomal pH was discussed under two hypotheses: (I) Endosomal pH increase, which is induced by proton pump, V-ATPase inactivation, causes the endosomal escape, or (II) the pH increase is not a cause of the endosomal escape, but a result of photoinduced destabilization of the endosomal membrane. In addition, the relationship of [Ca2+]i increase after photoirradiation with endosomal escape of TatU1A-PS/shRNA and the source of photoinduced [Ca2+]i increase in CPP mediated PCI was discussed. In chapter III, potential application of photoinduced cytosolic dispersion of RNA (PCDR) method to the study of cell differentiation mechanism in mouse embryos are discussed. In this study, PCDR was firstly confirmed in mouse embryos by using TatU1A-Alexa546/FAM shRNA. And then, photo-dependent knockdown of d1EGFP was demonstrated in four cell-stage embryos, indicating that spatial regulation of gene expression in mouse embryo was achieved by PCDR method. Finally, anti-aPKC shRNA, related to Par6-aPKC polarization, was introduced into a single cell of four-cell stage embryos and down regulation of PKC in early embryos development was briefly discussed.  論文審査結果の要旨 本研究では，光化学的内在化（PCI）と呼ばれる細胞内への物質導入法の機構解明，および PCI 手法のマウス胚細胞内への RNA デリバリーへの応用が行われた。   ①PCI とは，エンドサイトーシスにより細胞内に導入しようとした物質（生体分子や薬剤など）がエンドソーム内に閉じ込められてしまい，細胞質内に入らず機能しない問題に対する対策法の１つである。PCI 法では，目的物質を導入する際のキャリアとともに光増感剤を併用し，目的物質のエンドソーム内蓄積後に光照射を行うと，光増感剤が引き起こす反応により目的物質のエンドソームからの脱出が起こる。しかしながら，PCI 機構（光によりエンドソーム脱出が起こる機構）に関して，不明点は多い。この研究では，エンドソーム脱出に対する熱や pH や Ca2+イオン濃度の関与の有無について調べ，PCI 機構に関する新たな知見を得ることに成功した。 ②本研究では，PCI 機構にもとづいて働くことが分かっている「光応答性の RNA キャリア」を用いて，マウス胚細胞内への RNA の導入にも取り組んだ。狙いをつけて光照射した胚の中の１細胞だけに RNA を導入することに成功した。この手法により，胚の中の狙いをつけた細胞に対する miRNA の導入や，RNAi による特定の遺伝子のノックダウンなどが可能になるため，初期発生の研究に大いに役立つことが期待される。  これらの研究成果は，生命医用工学にとって有用な知見と方法論を提供するものだと考え，学位審査委員会は学位論文の内容，公聴会による発表内容等を総合的に判断し，本論文は博士（工学）に値するものと判定した。
