昆虫類の高次系統の再検討-内顎類全3目の内顎口の比較発生学的検討-著者 町田 龍一郎発行年 2012その他のタイトル Phylogenetic reconstruction of hexapod basalclades : Comparative embryological analyses ofentognathy formations all three entognathanconstituentsURL http://hdl.handle.net/2241/118327 様式Ｃ－１９  科学研究費助成事業（科学研究費補助金）研究成果報告書  平成 24 年 4 月 12 日現在                研究成果の概要（和文）：六脚類（広義の昆虫類）の高次系統「内顎類－外顎類システム」はこれまで広く支持されてきたが、近年、内顎類、欠尾類の単系統性を疑問視する系統仮説が提出されている。本研究は、内顎類 3目の重要な共有派生形質である内顎口の形成過程を厳密に比較することにより、内顎類の単系統性、3 目の系統関係を検証しようとするものである。結果、カマアシムシ目とトビムシ目の内顎口の相同性は支持されるが、カマアシムシ目/トビムシ目とコムシ目の内顎口の相同性は認められなかった。すなわち、欠尾類は妥当であるが、内顎類の単系統性は支持されない。  研究成果の概要（英文）：Recently, “Entognatha-Ectognatha System” has been challenged, and doubt has been cast on the interordinal relationships within and monophyly of Entognatha from multiple sources. Entognathy has been regarded as a most reliable “autapomorphy” of Entognatha. I compare the entognathy formations of Protura, Collembola and Diplura, examine the homology of their entognathies, and discuss the interordinal relationships and monophyly of Entognatha. Protura and Collembola are revealed to closely resemble each other in major points of entognathy formation, such as concerning: 1) the origin of mouth folds, which are formed by the extension and fusion of terga of intercalary and three gnathal segments, and 2) origin of the postgnathal region of entognathy, which is derived from labial terga medially migrated and united. However, the entognathy of Diplura is constructed in a different manner, which is characterized by: 1) the mouth fold is formed by the fusion of extended terga of intercalary and three gnathal segments and labial coxa, 2) the postgnathal region of entognathy is derived from labial subcoxae which medially migrate and unite into a form of postmentum, and 3) the rotation of labial appendages is involved in the entognathy formation. Thus, the homology of entognathy is well supported between Protura and Collembola, hence the monophyly of Ellipura may be supported. While the manner of entognathy formation in Diplura is revealed to be much different from in Protura and Collembola either, hence the homology of entognathy between Protura-Collembola and Diplura is not always verified, and the monophyly of Entognatha could not be substantiated.   交付決定額                                （金額単位：円）  直接経費 間接経費 合 計 2009 年度 1,300,000 390,000 1,690,000 2010 年度 1,200,000 360,000 1,560,000 2011 年度 1,100,000 330,000 1,430,000         総 計 3,600,000 1,080,000 4,680,000  機関番号：１２１０２ 研究種目：基盤研究（C） 研究期間：2009  ～ 2011  課題番号：２１５７００８９ 研究課題名（和文）昆虫類の高次系統の再検討－内顎類全３目の内顎口の比較発生学的検討－                      研究課題名（英文）Phylogenetic reconstruction of hexapod basal clades: Comparative embryological analyses of entognathy formations of all three entognathan constituents   研究代表者   町田 龍一郎（MACHIDA RYUICHIRO） 筑波大学・生命環境系・教授 研究者番号：５０１９９７２５      （   ）   研究者番号：     研究分野：動物系統分類学 科研費の分科・細目：基礎生物学・生物多様性・分類 キーワード：系統、昆虫比較発生学   １．研究開始当初の背景 研究代表者らは六脚類のベーサル・クレードの比較発生学的研究を一貫して行ない、「内顎類［＝欠尾類（＝カマアシムシ目＋トビムシ目）＋コムシ目］＋外顎類［イシノミ目＋双関節丘類（＝シミ目＋有翅昆虫類）］」との従来の六脚類（広義の昆虫類）の高次系統に対して、「胚膜の進化および胚と胚膜の機能分化の進化的変遷」との観点から、「カマアシムシ目＋《トビムシ目＋【コムシ目＋外顎類［イシノミ目＋双関節丘類（＝シミ目＋有翅昆虫類）］】》」との体系を導き、「内顎類」を棄却した。 確かに 、比較古生物学 （ Kukalová-Peck 1987）、頭部に関する機能形態学（Koch 2001）に「内顎類」のステータスを否定する研究はある。しかしながら、Hennig (1969) の提示した内顎類各目の「内顎口」の相同性に疑問を差し挟まない以上は、そのステータスは温存され、分子データとのコンバインによる系統構築でも同様である（Wheeler et al. 2001）。また、分子データからのみの系統構築においても（Luan et al. 2005）、支持確率は低いものの「内顎類」は生き続けている。 ここにあって、「内顎類」の検討のためには、内顎類各群間の「内顎口」の相同性を厳密に議論することが最重要の課題となる。そこで、申請者らは、まず、コムシ目ナガコムシ亜目の記載発生を行い（Ikeda & Machida 1998, 2001）、トビムシ目に関する先行研究との比較（Uemiya & Ando 1987）から、口褶（口器が被われているという「内顎口」たらしめる構造）の由来、内顎口形成にともなっての下唇付属肢の回転の有無という重要な点において、コムシ目とトビムシ目の内顎口形成は大きく異なることを明らかにし、これによりプランの異なる両目の「内顎口」の相同性は必ずしも支持されないとの結論を導いた。 さらに申請者らは初めてカマアシムシ目の発生学的研究に成功したが（Fukui & Machida 2006など）、この研究において、まだ予備的な段階ではあるが、興味深いデータを得た。すなわち、カマアシムシ目の内顎口は、コムシ目のそれとは異なり、口器の後方の被覆がコムシ目は下唇付属肢由来の後基板によるのに対し、カマアシムシ目では下唇背板の融合に由来する後頬板によりなされることが明らかになった。すなわち、カマアシムシ目とコムシ目両目に関しても、内顎口の相同性は必ずしも保証されない可能性が強くなってきた。  現在の六脚類の高次系統に関わる議論の争点は内顎類に関わるものであり、「内顎類」の唯一、最も確かとされてきた固有派生形質、「内顎口」を議論することが最優先であった。  ２．研究の目的 上述のような学術的背景から、「内顎類」の口器の比較形態学的検討が数多く試みられてきたが、分岐が極めて深い「内顎類」各群の特殊化した口器の比較は困難であり、決定打を欠く議論に終始してきた。このような行き詰った比較形態学的議論を打開するには、形成過程を検討して形態のプランの理解を目指す比較発生学的アプローチが最も有効である。本研究は、内顎類の唯一、最重要とされてきた、内顎類構成群の「共有派生形質」である「内顎口」の形成過程を比較検討することにより、上記の問題に切り込もうとするものである。  ３．研究の方法 内顎類全 3 目、すなわちカマアシムシ目、トビムシ目（フシトビムシ亜目とマルトビムシ亜目の全２亜目）、コムシ目（ナガコムシ亜目とハサミコムシ亜目の全２亜目）の内顎口形成を、種々の方法を駆使して、総合的かつ詳細に把握する。 （１） まず、各ステージの胚を卵から取り出しDAPI 染色し、蛍光実体顕微鏡で詳細に観察、各グループの内顎口形成を包括的に把握する。 （２） 固定胚を走査型電子顕微鏡用試料とし、内顎口形成を詳細な外部形態観察により理解する。 （３） 各グループの各ステージ胚を組織切片とし、光学顕微鏡、透過型電子顕微鏡観察を行い、内顎口形成過程を内部構造の観点からも把握する。 （４） 以上の観察を基に、各群の内顎口のグラウンドプランを構築する。それらのグラウンドプランを、異同を厳密に議論しながら比較することにより、系統学的議論を発展させる。  ４．研究成果 内顎類全 3 目、すなわち、カマアシムシ目、トビムシ目（フシトビムシ亜目とマルトビムシ亜目の全２亜目）、コムシ目（ナガコムシ亜目とハサミコムシ亜目の全２亜目）の内顎口形成を、種々の方法を駆使して、総合的かつ詳細に観察した。その結果、各群の内顎口のグラウンドプランを下記のように理解することができた。 （1） トビムシ目において、口褶は間挿・大顎・小顎・下唇体節背板に由来することが明らかになった。左右の下唇亜基節の正中での融合で形成された後基板は、左右の下唇背板の過剰な拡張により完全に覆われる。すなわち、  トビムシ目の内顎口の後方部は下唇背板由来である。以上の結果は主にフシトビムシ亜目から導かれたものであるが、一方のマルトビムシ亜目に関する断片的な観察結果に矛盾する点は認められない。いくつかのトビムシ目の内顎口形成に関する先行研究はいずれも断片的かつ解像度の極めて低いものであったが、本研究はこれらのあやふやない理解を一新するものである。 （2） カマアシムシ目の内顎口形成についても詳細に理解することができた。その結果、カマアシムシ目の内顎口は、口褶および内顎口後方部の由来において、トビムシ目のそれに本質的に一致することが明らかになった。すなわち、カマアシムシ目の内顎口とトビムシ目の内顎口は相同である可能性が極めて高い。 （3） 一方で、コムシ目の内顎口のグラウンドプランは、トビムシ目、カマアシムシ目のそれとは極めて異なることが明らかになった。すなわち、間挿・大顎・小顎・下唇体節背板に加え、下唇基節が口褶の形成に参加、また、内顎口後方は後基板にのみ由来する。さらに、内顎口形成に 90度の下唇付属肢の回転が伴う点も、カマアシムシ目、トビムシ目と大きく異なっている。上記の相違は、カマアシムシ目－トビムシ目の内顎口とコムシ目のそれを相同視するにはあまりに大きいものであり、両者間の相同性は支持されない。 （4） 以上から、カマアシムシ目とトビムシ目の近縁性は強く示唆され、両目からなる欠尾類の単系統性は強く支持される。一方で、欠尾類とコムシ目の姉妹群関係を前提とする内顎類の単系統性は支持されない。六脚類のベーサル・クレードの従来の系統学的理解に対し、例えば、私たちが比較発生学的立場から提出してきた、「カマアシムシ目＋《トビムシ目＋【コムシ目＋外顎類［イシノミ目＋双関節丘類（＝シミ目＋有翅昆虫類）］】》」との系統仮説の説得力はさらに強化される。  ５．主な発表論文等 （研究代表者、研究分担者及び連携研究者には下線）  〔雑誌論文〕（計３９件） ① Dallai, R., D.
