  Abstract The proteinogenic amino acid L-serine has manifold functions in plant metabolism and is indispensable for plant growth and development. Plants possess at least two main pathways for the biosynthesis of serine: the photorespiration, which takes place in the photosynthetic tissue of the plant, and the phosphorylated pathway of serine biosynthesis (PPSB). The latter was postulated to be involved in serine supply to heterotrophic plant tissue. The photorespiration is considered as the most important pathway, as it produces vast amounts of serine. However, despite a functional photorespiratory pathway and unchanged steady-state levels of serine, plants deficient in the PPSB exhibit a strong impairment in plant root growth. Moreover, silencing of PGDH1, the rate limiting enzyme of this pathway, results in a substantial perturbation of plant metabolism, especially in nitrogen assimilation, as indicated by amino acid accumulation and increased ammonium content.  However, the reason for the metabolic and morphological alterations in PPSB-deficient plants are not yet fully elucidated. Therefore, this study aimed at investigating the role of the PPSB in plant serine homeostasis and in plant metabolism in general. To do so, we dissected the contribution that each biosynthesis pathway has in plant serine supply by suppressing the photorespiration and the PPSB individually and simultaneously. The consequences on plant growth and metabolism were examined by phenotypical characterization, metabolite flux analysis using heavy isotopy labeling and mass spectrometry-based metabolite profiling. The results of this study clearly support the important role of the PPSB for plant growth and metabolism. Furthermore, we could show that the PPSB is even more important for maintaining serine homeostasis than photorespiration. Although the predominant function of the PPSB lies in the provision of serine to root tissue, it also feeds serine to the shoot. Especially the meristematic tissues in shoot and root seem to depend on serine supply via the PPSB. Moreover, tracer studies indicated that PPSB-derived serine is consumed by all major serine-depending pathways, such as the synthesis of DNA, proteins, lipids and polar metabolites. Furthermore, we showed that deficiency in PPSB-derived serine leads to a significant impairment of protein biosynthesis.  In accordance with the central role of the PPSB in plants, PGDH1-deficiency results in a high amino acid phenotype. This study revealed that amino acid accumulation in these plants clearly results from serine deficiency and is at least partially associated with impaired protein biosynthesis. However, stable isotope labeling experiments illustrated that the amino acid phenotype is also caused by increased nitrogen assimilation, especially in root tissue. In addition, we found that activity and flux mode of the respiratory metabolism is altered in PGDH1-silenced plants, most likely as a consequence of amino acid accumulation and elevated nitrogen assimilation.    Zusammenfassung Die proteinogene Aminosäure L-Serine besitzt vielfältige Funktionen im pflanzlichen Stoffwechsel und ist unerlässlich für das pflanzliche Wachstum und die Entwicklung. Pflanzen verfügen über mindestens zwei Stoffwechselwege für die Synthese von Serin: die Photorespiration, welche im photosynthetisch aktiven Gewebe stattfindet, sowie den phosphorylierten Stoffwechselweg der Serinsynthese (engl. phosphorylated pathway of serine biosynthesis, PPSB). Für den Letzteren wurde eine Beteiligung in der Serinversorgung von heterotrophem Gewebe postuliert. Die Photorespiration gilt als wichtigster Serinbiosyntheseweg, da sie große Mengen dieser Aminosäure produziert. Jedoch zeigen Pflanzen mit fehlendem PPSB trotz einer funktionierenden Photorespiration und unveränderten Seringehalten eine starke Beeinträchtigung des Wachstums. Darüber hinaus führt eine Reduzierung der Aktivität von PGDH1, dem geschwindigkeitsbestimmenden Enzym dieses Stoffwechselweges, zu einer erheblichen Störung des pflanzlichen Metabolismus, insbesondere der Stickstoffassimilierung, wie aus Aminosäurenakkumulation und erhöhtem Ammoniumgehalt ersichtlich wird.  Die Gründe für diese metabolischen und morphologischen Veränderungen in PPSB-defizienten Pflanzen sind jedoch noch nicht vollständig ergründet. Daher bestand das Ziel dieser Arbeit in der genauen Untersuchung der Aufgabe des PPSBs in der pflanzlichen Serinhomöostase sowie allgemein im pflanzlichen Metabolismus. Zu diesem Zweck analysierten wir den Beitrag der einzelnen Biosynthesewege in der pflanzlichen Serinversorgung, indem die Photorespiration und der PPSB einzeln und gemeinsam ausgeschaltet wurden. Die Auswirkungen auf das pflanzliche Wachstum und den Metabolismus wurden mittels phänotypischer Charakterisierungen, Analyse des Metabolitenflusses durch Isotopenmarkierung und massenspektrometrischer Metabolituntersuchungen erforscht.  Die Ergebnisse dieser Studie bekräftigten eindeutig die wichtige Rolle des PPSBs für das Wachstum und den Stoffwechsel der Pflanze. Darüber hinaus konnten wir belegen, dass der PPSB für die Aufrechterhaltung der Serinhomöostase sogar von größerer Bedeutung ist als die Photorespiration. Obwohl die Hauptfunktion des PPSBs in der Serinversorgung des Wurzelgewebes liegt, stellt dieser Stoffwechselweg auch Serin für den Spross bereit. Besonders das meristematische Gewebe von Spross und Wurzel scheint auf die Serinversorgung durch den PPSB angewiesen zu sein. Darüber hinaus wiesen Isotopen-Studien darauf hin, dass Serin aus dem PPSB von allen serinabhängigen Hauptstoffwechselwegen, wie zum Beispiel der Synthese von DNS, Proteinen, Lipiden und polaren Metaboliten, genutzt wird. Zudem konnten wir zeigen, dass ein Mangel an Serin aus dem PPSB zu einer erheblichen Beeinträchtigung der Proteinbiosynthese führt.      Im Einklang mit der zentralen Rolle des PPSBs in der Pflanze, führt ein PGDH1-Mangel zu einem starken Aminosäure-Phänotyp. Diese Studie konnte nachweisen, dass die Aminosäureakkumulation eindeutig auf einem Serinmangel beruht und zumindest teilweise mit der beeinträchtigten Proteinbiosynthese zusammenhängt. Allerdings zeigten Experimente mittels Isotopenmarkierung, dass der Aminosäurephänotyp auch durch eine erhöhte Stickstoffassimilierung, vor allem im Wurzelgewebe, hervorgerufen wird. Ferner konnten wir eine Veränderung der Aktivität und des Flussmodus des respiratorischen Metabolismus in PGDH1-defizienten Pflanzen nachweisen. Dies ist wahrscheinlich eine Folge der Aminosäurenakkumulation und der erhöhten Stickstoffassimilierung.
