The ubiquitin ligase activity of cIAP1 and cIAP2 controls  cell death-dependent and -independent tissue inflammation  Fabian Schorn  Abstract The E3 ubiquitin ligases cIAP1 and 2 are crucial regulators of cell survival and pro-inflammatory signaling by inducing the formation of a ubiquitin-dependent scaffolding network leading to canonical NFkB and MAPK signaling. Additionally, cIAP1/2 have been shown to negatively regulate RIPK1-mediated cell death and activation of the non-canonical NF-kB signaling pathway through ubiquitin-dependent degradation of NIK.  Previous work using genetic mouse models or chemical depletion have been used to investigate the physiological role of cIAP1/2 in vivo and in vitro. Whereas single deficient mice were viable, cIAP1/2 double knockout animals died during embryogenesis indicating vital, but redundant functions during development. Also, mice with a single E3 ligase deficiency were vastly normal yet revealed potential cell type specific differences. In order to investigate the physiological relevance of cIAP1/2 E3 ligase activity, we established a new mouse model simultaneously expressing cIAP1/2 E3 ligase deficient variants (cIAP1/2MutR/MutR) using the CRISPR/Cas9 approach. We introduced short deletions in the RING domain of cIAP1 and cIAP2 which abrogated the E3 ligase function and found that homozygous expression of cIAP1/2 E3 ligase mutants resulted in embryonic lethality at E10.5 due to cardiovascular defects and excessive endothelial cell death. Deletion of either TNFR1 or expression of RIPK1 kinase-dead rescued embryonic development. Yet, TNFR1Ko/Ko cIAP1/2MutR/MutR and RIPK1D138N/D138N cIAP1/2MutR/MutR mice showed premature lethality which could not be rescued when necroptosis was additionally blocked in TNFR1Ko/Ko RIPK3Ko/Ko cIAP1/2MutR/MutR mice. Histological analyses in these mice revealed systemic inflammation characterized by immune cell infiltration, upregulated pro-inflammatory cytokine levels and increased cell death. Strikingly, mice with a combined TNFR1Ko/Ko RIPK1D138N/D138N cIAP1/2MutR/MutR background showed significantly increased survival but still succumbed due to systemic multi-tissue inflammation. This late-stage phenotype might be triggered by NIK-dependent non-canonical NF-kB and immune cell driven hyperinflammation which could not be blocked by ablation of TNFR1 or RIPK1 kinase activity.  This study provides new insights into the physiological role of the cIAP1/2 E3 ligase activity to safeguard embryonic development and to prevent inflammatory tissue destruction in adult tissues.   Zusammenfassung Die E3 Ubiquitin Ligasen cIAP1 und cIAP2 (cIAP1/2) sind wichtige Regulatoren innerhalb verschiedener Signalkaskaden. Durch ihre E3 Ubiquitin Ligase Aktivität induzieren cIAP1/2 die Ausbildung eines Ubiquitinnetzwerkes, das als Bindungsplattform dient, und letztendlich für die Aktivierung der kanonischen NF-kB und MAPK Signalwege sorgt. Außerdem wurde gezeigt, dass cIAP1/2 negative Regulatoren RIPK1-induzierten Zelltods und NIK-abhängiger Aktivierung des nicht-kanonischen NF-kB Signalweges sind. Knockout-Mäuse (KO) und chemische Inhibierung wurden verwendet, um die Role von cIAP1/2 in vivo und in vitro zu untersuchen. Mäuse mit einer einzelnen cIAP1 bzw. cIAP2 KOs entwickelten sich normal und zeigten keine phänotypischen Auffälligkeiten, wohingegen cIAP1/2 Doppel-KO-Mäuse während der Embryonalentwicklung an Tag 10.5 (E10.5) verstarben. Mäuse mit einzelnen E3 Ligase-inaktiven Mutationen zeigten keine besonderen Auffälligkeiten, teilweise wurden Zelltyp-spezifische Aktivitätsunterschiede festgestellt.  In der vorliegenden Arbeit haben wir ein neues Mausmodel etabliert, das es ermöglicht die Funktionsweise von cIAP1/2 besser zu verstehen. Durch die CRISPR/Cas9 Methode wurden parallel inaktivierende Mutationen in die RING Domäne von cIAP1/2 eingeführt, welche die E3 Ligase Aktivität zerstören. Mäuse, die homozygot für die E3 Ligase Mutanten sind (cIAP1/2MutR/MutR), sterben während der Embryonalentwicklung an Tag 10.5 (E10.5), vermutlich ausgelöst durch den Tod von Endothelzellen und kardiovaskulären Defekten. Eine Deletion von TNFR1, oder die Expression einer Kinase-inaktiven RIPK1 Variante verhindert den vorzeitigen Tod. Dennoch zeigen TNFR1Ko/Ko cIAP1/2MutR/MutR und RIPK1D138N/D138N cIAP1/2MutR/MutR Tiere frühzeitige Letalität. Eine RIPK3 Deletion, zu Necroptose Blockade, brachte keinen Überlebensvorteil in TNFR1Ko/Ko RIPK3Ko/Ko cIAP1/2MutR/MutR Tieren. Histologische Untersuchungen zeigten eine systemische Entzündung, die sich durch Immunzellinfiltrate, hochregulierte Zytokinlevel und erhöhten Zelltod auszeichnete. TNFR1Ko/Ko RIPK1D138N/D138N cIAP1/2MutR/MutR Tiere zeigten ein deutlich verlängertes Überleben und dennoch entwickelten auch diese Tiere eine systemische Entzündung, die zum vorzeitigen Tod führte. Dieser verzögerte Phänotyp könnte durch unkontrollierte nicht-kanonischen NF-kB Aktivierung und eine Immunzell-abhängige Entzündung hervorgerufen werden. Zusammengefasst zeigt die Studie die Wichtigkeit der cIAP1/2 Ligase Aktivität während der Embryonalentwicklung und ihre physiologische Relevanz in adulten Geweben, um unkontrollierten Zelltod und Entzündungsreaktionen zu blockieren.
