However, many species are threatened by overharvesting and habitat loss, and their conservation is urgent. In addition, orchids are well-known in the global floriculture industry. Orchidaceae plants have developed a unique symbiotic relationship with mycorrhizal fungi for nutrient supply. Mycorrhizal associations can benefit orchid plants, including improved vegetative and reproductive growth. However, knowledge of mycorrhizal associations in wild and cultivated orchids is still limited.   This study investigated the mycorrhizal association of a wild orchid, Phalaenopsis japonica, distributed in Japan. Mycorrhizal fungi were molecularly identified using a total of 32 individuals from 4 sites, revealing that this species is mainly associated with a specific lineage belonging to the Basidiomycete Ceratobasidiaceae. A symbiotic culture experiment showed that the particular fungi could promote both seed germination and seedling growth, while the other fungi promoted seed germination less and only seedling growth. This study investigated the mycorrhizal association of Phalaenopsis species for the first time, providing valuable insights into the conservation of wild orchids.   This study also investigated the mycorrhizal association of cultivated Dendrobium hybrid. A single cultivar from three nurseries was evaluated for mycorrhizal colonization. Microscopic observation showed that all 13 plants examined were highly colonized by mycorrhizal fungi. The mycorrhizal communities were assessed by metabarcoding analysis. The results showed that the fungal communities were largely different among the three nurseries, suggesting that the culture environment significantly affects the mycorrhizal association. The mycorrhizal fungi isolated from roots promoted the seed germination of three Dendrobium species and the seedling growth of Dendrobium moniliforme. These results suggest that the mycorrhizal fungi associated with cultivated orchids have promotive effects on orchid germination and growth. This  is the first comprehensive study of mycorrhizal association in a horticultural orchid hybrid and provides valuable insights for commercial production.
