circRNA-miRNA-mRNA调控网络的挖掘以筛选缺血性脑卒中的潜在治疗药物

Abstract
目的挖掘缺血性脑卒中潜在的circRNA-miRNA-mRNA调控网络及其相应的治疗药物。方法利用GEO数据库发布的数据并选择研究大脑中动脉栓塞(MCAO)小鼠RNA表达水平的3个数据集GSE115697、GSE51586和GSE137482,使用R中的Limma包来筛选差异表达的mRNA(DEmRNA)、DEcircRNA和DEmiRNA。采用MiRWalk、StarBase等多种生物信息学方法结合的综合策略来挖掘与脑卒中相关的circRNA-miRNA-mRNA网络,由Cytoscape软件完成核心子网络的筛选和可视化输出。通过基因本体(GO)富集分析及京都基因与基因组百科全书(KEGG)通路分析研究ceRNA网络的功能,以富集超过10个基因且假发现率(FDR)<0.05的通路认为具有统计学意义。最后使用药物基因组学数据库connectivity map(CMap)筛选潜在的脑卒中治疗药物,并对野生型小鼠构建MCAO模型验证药物治疗效果。结果共筛选得出1 249个DEmRNA、294个DEmiRNA和70个DEcircRNA,并发现3个circRNA-miRNA-mRNA调控网络可能与缺血性脑卒中密切相关。根据上述结果筛选得出Austricine、Metyrapone和Desoxypeganine这3种最有可能逆转由脑卒中相关ceRNA网络引起的改变的药物,并以Metyrapone为例验证所筛选药物对缺血性脑卒中确实可以改善术后第一(P=0.001 4)、二天(P=0.016 1)的神经功能损伤和减少脑梗死体积(P=0.004 9),发挥脑保护作用。结论本研究从circRNA-miRNA-mRNA网络视角为缺血性脑卒中的发病机制提供了新的见解,本文预测得出的3种药物为缺血性脑卒中的治疗方法提供了新的线索
