       Quality Control Processes in  IMS Protein Import Inaugural-Dissertation zur Erlangung des Doktorgrades der Mathematisch-Naturwissenschaftlichen Fakultät der Universität zu Köln  vorgelegt von Markus Andreas Habich aus Simmern  Köln, 2019      Zusammenfassung Die Bildung von Disulfidbrücken im mitochondrial Intermembranraum (IMS) ist ein essentieller Prozess. Die Oxidoreduktase CHCHD4/Mia40 ist das Schlüsselenzym dieser Reaktion und koppelt die Oxidation von Substratproteinen mit deren Import. Obwohl bisherige Studien viel zum mechanistischen Verständnis der Disulfidbrückenbildung im IMS beitrugen, blieb bisher unklar, wie diese mit dem Import von Substraten gekoppelt ist. Im ersten Teil dieser Arbeit zeigen wir, dass Substrate während ihres Imports einen metastabilen disulfidgebundenen Komplex mit CHCHD4 ausbilden und dass dieser wichtig für deren Import ist. Unsere Ergebnisse deuten darauf hin, dass auf Ebene dieses Komplexes über das Schicksal des Substrates entschieden wird. Kommt es nach Komplexbildung nicht zur produktiven Oxidation und Import des Substrates, wird der Komplex über eine Redoxqualitätskontrolle aufgelöst und das Substrat abgebaut. Wir zeigen, dass, falls die Bildung dieses Komplexes von vornerein verhindert wird oder nach Komplexbildung keine erfolgreiche Oxidation erfolgt, Substrate ins Zytosol freigesetzt und durch das Proteasom abgebaut werden können. Somit ist im Mechanismus des vektoriellen Imports, ein Kontrollschritt implementiert, der die Auflösung von unproduktiven Substratinteraktionen erlaubt. Die Erkenntnisse dieser Arbeit bieten Aufschluss über das Schicksal von verschiedenen krankheitsassoziierten CHCHD4-Substratmutanten, welche in ihrer mitochondrialen Akkumulation beeinträchtigt sind.  Bisher ist weiterhin unklar, welche zytosolischen Prozesse CHCHD4-Substrate vor ihrem Import in Mitochondrien durchlaufen. Das kürzlich von unserer Gruppe identifizierte unkonventionelle CHCHD4-Substrat AK2 erwies sich als besonders instabil im Zytosol. Im zweiten Teil dieser Arbeit zeigen wir, dass die Stabilität von AK2 durch einen zytosolischen Qualitätskontrollschritt reguliert wird. Die N-terminale Prozessierung von AK2 durch die zytosolischen Proteasen DPP8/9 bewirkt seinen zytosolischen Abbau durch das Proteasom. Unsere Ergebnisse deuten darauf hin, dass die Modulation dieses Schrittes die zytosolische Akkumulation und Aktivität von AK2 erlaubt. Weiterhin ergab die Analyse des MitoCarta2.0-Proteoms hinsichtlich möglicher Schnittstellen, dass mehrere weitere mitochondriale Proteine Substrate für DPP8/9 sein könnten. Dies deutet darauf hin, dass die N-terminale Prozessierung durch DPP8/9 eine generelle Rolle in der mitochondrialen Biogenese einnehmen könnte. Unsere Daten zeigen, dass auch eine krankheitsassoziierte Mutante der Komplex-I-Untereinheit NDUFB10 durch diesen Weg reguliert wird, was zu unserem Verständnis des Substratschicksals im Kontext von humanen Krankheiten beiträgt.        Abstract Disulfide formation and import into the mitochondrial intermembrane space (IMS) is an essential process. It is catalyzed by the disulfide relay machinery which couples oxidation of proteins to their import. The key enzyme of this machinery is the oxidoreductase and chaperone CHCHD4/Mia40. While mechanisms of substrate oxidation are well-understood, the question of how it is coupled to protein import remains open. In the first part of this thesis, we show that incoming substrates initiate a metastable disulfide-linked complex with CHCHD4 that is critical for their import. We propose that this disulfide-linked complex is at crossroads to productive oxidation and import or to a redox quality control step that mediates resolving of the complex and substrate degradation. We show that if this complex is not followed by productive oxidation, or its formation is prevented all together, substrates become released to the cytosol and degraded by the proteasome. Therefore, the disulfide relay itself surveils, based on the vectorial nature of IMS import, the folding-competence of substrates at the level of the disulfide-linked CHCHD4-substrate complex. These findings help to understand the fate of disease-associated mutant variants of CHCHD4 substrates that are compromised in their mitochondrial accumulation.  So far, knowledge on processes occurring in the cytosol before translocation of disulfide relay substrates is scarce. When our group identified and characterized the unconventional substrate AK2, we found that it is highly unstable in the cytosol. In the second part of this thesis, we show that AK2 is subject to a cytosolic quality control step which regulates AK2-stability in the cytosol. N-terminal processing of AK2 by the cytosolic proteases DPP8/9 renders AK2 prone to degradation by the proteasome. Our results indicate that modulation of this step allows for cytosolic accumulation and activity of AK2. Furthermore, screening the MitoCarta2.0 proteome for potential cleavage sites indicated that numerous mitochondrial proteins might be substrates of DPP8/9 suggesting a broad role of N-terminal processing in mitochondrial biogenesis. Our data indicate that also a disease-associated mutant of the complex I subunit NDUFB10 is subject to this quality control pathway thereby providing insight on substrate fate in human disease conditions.
