Abstract  Phosphorus is an essential nutrient for plant growth. The pool of plant available phosphorus in the soil (phosphate) is usually limited. As a consequence plants adjust their metabolism and morphology by mounting the phosphate starvation response (PSR). PSR is regulated by a complex genetic network, which is interconnected by signals acting at the local and systemic level. PSR signals include phosphate, phytohormones, microRNAs and sucrose, which is the one of the most important systemic PSR signals. To increase our understanding of the plant PSR and its dependency on sucrose and/or photoassimilates, a transcriptome analysis of A. thaliana seedlings growing in contrasting sucrose, phosphate and light conditions was performed. Differentially expressed genes (DEG) analysis allowed to identify the genes responding to contrasting phosphate treatments in presence or absence of photoassimilates and externally supplied sucrose. A small set of P-responsive genes was found in seedlings grown in the dark without sucrose supplementation, suggesting that there can be a light/sucrose-independent PSR signaling. Sucrose greatly enhanced the plant response to low phosphate, as shown by the increased expression of genes encoding P transporters, phosphatases, transcription factors, among others. The expression pattern of DEG in response to phosphate shared similarity with A. thaliana responses to biotic stresses like pathogen attack. This suggests that the plant PSR could affect plant-microbe interactions.  Contrasting with most land plants, A. thaliana has lost the ability to establish the arbuscular mycorrhizal (AM) symbiosis, a partnership considered to be essential for P scavenging from soil. In mycorrhizal plants the colonization of plant roots by AM fungi strongly depends on plant PSR and P status. Still, not much is known about the impact of P and the PSR on other plant-fungus interactions. Hence, the second part of this thesis addressed the questions if and how the plant PSR influences the root-associated fungal microbiome in the non-mycorrhizal plant A. thaliana. P supply to the soil changed root- associated fungal communities, but not bulk soil communities, implying that the modification of the plant PSR was the main driver of these changes. Fungi colonized P-starved and P-replete plants differently; this was observable already at the taxonomical order level, as Helotiales, Xylariales and Hypocreales were more abundant in P-starved plants. These fungal communities differences between P-starved and P-replete plants could not be observed when the plant PSR was changed systemically via the shoot, suggesting a major effect of local/root responses to P on plant-fungus interactions. Moreover, A. thaliana mutants affected in their PSR exhibited a altered root-associated fungal communities.   Zusammenfassung  Phosphor ist ein essenzieller Nährstoff für das Pflanzenwachstum. Der pflanzenverfügbare Phosphor in der Erde (Phosphat) ist häufig limitiert. In Folge dessen passen Pflanzen ihren Metabolismus und ihre Morphologie durch eine Phosphatmangelantwort (phosphate starvation response, PSR) an. PSR wird durch ein komplexes genetisches Netzwerk reguliert, welches durch Signale auf lokaler und systemischer Ebene verknüpft ist. Diese Signale sind Phosphat, Phytohormone, microRNAs und Saccharose, welches eins der wichtigsten systemischen Signale in der PSR ist.  Um den Zusammenhang der pflanzlichen PSR und ihre Abhängigkeit von Saccharose und Licht besser zu verstehen, wurde eine Transkriptomanalyse von A. thaliana Keimlingen in kontrastierenden Saccharose-, Phosphat- und Lichtbedingungen durchgeführt. Eine Analyse von differenziell exprimierten Genen (differentially expressed genes, DEG) erlaubte es Gene zu identifizieren, welche unterschiedlich auf die kontrastierenden Phosphatbedingungen in An- und Abwesenheit von Photoassimilaten und extern zugefütterter Saccharose reagierten. Es wurden P-assozierte Gene in Keimlingen gefunden, welche im Dunkeln ohne Saccharosezugabe gewachsen sind, dies deutet darauf hin, dass es von Licht/Saccharose unabhängige Signale in der PSR gibt. Saccharose steigert die Antwort auf Phosphatmangel durch die vermehrte Expression von Genen, welche unter anderem für P Transporter, Phosphatasen und Transkriptionsfaktoren kodieren. Das Expressionsmuster der DEG in der Phosphatantwort ist ähnlich zu der Reaktion auf biotischen Stress wie Pathogenbefall in A. thaliana. Daraus lässt sich schließen, dass die pflanzliche PSR die Interaktion von Pflanzen und Mikroben beeinflussen könnte.  Im Unterschied zu vielen anderen Landpflanzen hat A. thaliana die Fähigkeit verloren eine arbuskuläre Mycorrhiza (AM) einzugehen, diese Symbiose wird als essenziell für die Gewinnung von P aus der Erde betrachtet. Der Kolonisierungsgrad der Pflanzenwurzeln durch AM Pilze ist abhängig von der pflanzlichen PSR und dem P Status. Trotz allem ist bis jetzt noch nicht viel bekannt über die Auswirkung von P und PSR auf andere Interaktionen zwischen Pflanzen und Pilzen. Deshalb adressiert der zweite Teil dieser Arbeit die Frage, ob und wie die pflanzliche PSR das pilzliche Mikrobiom, welches mit der Wurzel assoziert ist, in der nicht zu mycorrhizierbaren Pflanze A. thaliana beeinflusst. Die Versorgung mit P veränderte die mit der Wurzel-assozierte Pilzgemeinschaft, aber nicht die Pilzgemeinschaft in der Bodenmasse, dies impliziert, dass die Modifikation der pflanzlichen PSR der Hauptgrund für diese Veränderung ist. Des Weiteren kolonisierten Pilze P-gemangelte und P-gesättigte Pflanzen unterschiedlich. Dies konnte schon auf der Ebene der taxonomischen Ordnung beobachtet werden, da Helotiales, Xylariales und Hypocreales gehäuft in P-gemangelten Pflanzen vorkamen. Diese Unterschiede in der pilzlichen Gemeinschaft zwischen P-gemangelten und P-gesättigten Pflanzen konnten nicht beobachtet werden, wenn die pflanzliche PSR systemisch durch den Spross verändert wurde. Dies weist auf einen großen Einfluss der lokalen sowie der Wurzelantwort durch P auf die Interaktion zwischen Pilz und Pflanze hin. Zudem zeigten A. thaliana Mutanten, welche in der PSR beeinträchtigt waren, eine veränderte mit der Wurzel-assozierte pilzliche Artengemeinschaft.
