Abstract  Light is one of the most important environmental factors which regulate developmental and metabolic responses such as seedling de-etiolation, phototropism and the biosynthesis of anthocyanins. Anthocyanins are natural pigments that accumulate only in light-grown Arabidopsis plants and this accumulation is further enhanced by a number of factors, for example sucrose. The biosynthesis of anthocyanins is regulated by transcription factors that induce the expression of structural genes that code for enzymes in the biosynthesis pathway. These transcription factors include HY5 and a complex consisting of a WD-repeat, bHLH and MYB proteins. In the dark, light responses including light-induced anthocyanin production is suppressed by the activities of the CONSTITUTIVELY PHOTOMORPHOGENIC1/SUPPRESSOR OF PHYA-105 (COP1/SPA) complex, a tetrameric complex consisting of two COP1 and two SPA proteins. In dark-grown plants, this complex acts as a CUL4/DDB1-based ubiquitin ligase to ubiquitinate transcription factors that are required for the light response, thereby targeting them for degradation in the 26S proteasome. To enhance our understanding of the mechanisms involved in light-dependent sucrose-induced anthocyanin biosynthesis in Arabidopsis thaliana, the transcriptomic analyses on the basis of Illumina sequencing were performed. In this order the multifactorial RNAseq experiment treating seedlings with/without light, with/without sucrose and with/without phosphate was planned. As a genetic control for signaling activity rather than energy supply various signaling mutants were included besides the wild type (e.g. cop1-6, hy5-215). It was investigated how light signaling, on its own as well as in combination with sucrose signaling, modulates the expression of anthocyanin regulatory as well as structural genes. Among the anthocyanin regulatory genes the R2R3-MYB transcription factors are found as major regulators of the anthocyanin biosynthesis pathway. In this study, a novel R2R3 MYB gene was isolated and characterized in Arabis alpina. This MYB gene is closely related to the Arabidopsis thaliana AtPAP1 and PAP2, and has been designated as AaPAP. Ectopic expression of AaPAP in Arabidopsis thaliana significantly enhanced the anthocyanin biosynthesis and accumulation. Yeast two-hybrid assay showed that AaPAP was capable of interacting with Arabidopsis bHLH regulator of the anthocyanin biosynthetic pathway. These results suggest that EsAN2 is involved in regulation of the anthocyanin biosynthesis in Arabis alpina. In this study, the details of the interaction between the WD40 domain of COP1 and SPA and their interaction partners were characterized. By point mutations in COP1- interaction motif present in Arabidopsis PAP2 the details of the interaction between PAP2 and with WD40 repeat domain of COP1 and SPA protein was analyzed. The new domain interacting motif present in PAP2 and in the new COP1/SPA-interacting proteins was defined. Furthermore, by site directed mutagenesis approach the contribution of these domains in protein stability both in the dark as well in the light were studied.  Zusammenfassung  Das Sonnenlicht ist einer der wichtigsten Umweltfaktoren, welches erheblichen Einfluss auf die metabolischen Prozesse und die Entwicklung der Pflanzen nimmt. Beispiele für eine Licht abhängige Reaktion ist die De-etiolierung von Keimlingen, der Phototropismus oder die Synthese von Anthocyanen. Hierbei sind Anthocyane natürliche vorkommende Pigmente, wie sie ausschließlich in Licht angezognenen Arabidopsis Pflanzen vorkommen. Die Stärke der Anthocyan Produktion kann jedoch durch weitere Faktoren, wie die Zugabe von Saccharose, beeinflusst werden. Ferner ist die Synthese der Anthocyane eng durch Transkriptionsfaktoren reguliert, welche über die Expression der Gene für die Biosynthese Einfluss nehmen. Diese Transkriptionsfaktoren beinhalten HY5-, bHLH- und MYB-Proteine, sowie ein weiterer Komplex aus WD- Repeat Proteinen. In Dunkelheit wird die Lichtinduzierte Synthese von Anthocyanen durch die Aktivität eines Supressor Protein Komplex, dem CONSTITUTIVELY PHOTOMORPHOGENIC1/SUPPRESSOR OF PHYA-105 (COP1/SPA) Komplex reguliert.   Der tetramerische Komplex besteht aus zwei COP1 und zwei SPA Proteinen. Bei in Dunkelheit gewachsenen Pflanzen, fungiert der COP1/SPA Komplex als CUL4/DDB1-basierte Ubiquitin Ligase um Transkriptionsfaktoren zu ubiquitinieren, die für die Licht- Antwort verantwortlich sind und markiert diese für den Abbau im 26S Proteasom. Die Transkriptionelle Regulation, post-translationale Modifizierungen und Abbau von Transkriptionsfaktoren sind wichtig in der Licht-regulierten Kontrolle der Pflanzenentwicklung. Um unser Verständnis der Mechanismen der lichtabhängigen Saccharose-induzierten Anthocyanin-Biosynthese in Arabidopsis thaliana zu verbessern, wurden die transkriptomischen Analysen auf der Basis der Illumina-Sequenzierung durchgeführt. Aus diesem Grund wurde ein multifaktorielles RNAseq-Experiment geplant, bei dem Keimlinge mit und ohne Licht, sowie mit und ohne Saccharose, als auch mit und ohne Phosphat behandelt wurden. Als genetische Kontrolle für die Aktivität des Anthocyanin Signalweges und nicht dem Signalweg für die Energieversorgung wurden neben dem Wildtyp verschiedene Mutanten (z. B. cop1, hy5) mit einbezogen. Im Speziellen wurde untersucht, wie der Lichtsignalweg, in Kombination mit dem Saccharose Signalweg, die Expression von Genen der Anthocyanin Synthese zu regulieren und modulieren vermag. Unter den Anthocyanin-regulatorischen Genen werden die R2R3-MYB Transkriptionsfaktoren als Hauptregulatoren des Anthocyanin-Biosynthesewegs beschrieben. In dieser Studie wurde ein neuartiges R2R3-MYB-Gen isoliert und in Arabis alpina charakterisiert. Dieses MYB-Gen ist eng mit den Genen PAP1 und PAP2 aus Arabidopsis thaliana verwandt und wird im Folgenden als AaPAP bezeichnet. Die ectopische Expression von AaPAP in Arabidopsis thaliana erhöhte deutlich die Anthocyanin-Biosynthese und Anthocyanin Akkumulation. Der Hefe-Zwei-Hybrid-Assay zeigte, dass AaPAP mit den bHLH Regulatoren des Anthocyanin-Biosynthesewegs in Arabidopsis in Wechselwirkung tritt. Die gewonnenen Ergebnisse deuten darauf hin, dass EsAN2 an der Regulation der Anthocyanin- Biosynthese in Arabis alpina beteiligt ist.  In der vorliegenden Arbeit wurden die Details der Interaktion zwischen der WD40- Domäne von COP1 und SPA und ihren Interaktionspartnern charakterisiert. Durch Punkt- Mutationen im COP1-Interaktionsmotiv in Arabidopsis PAP2 wurden die Details der Wechselwirkung zwischen PAP2 und WD40-Repeat Domäne von COP1 und SPA-Protein analysiert. Hierdurch konnte ein neues Motiv für die PAP2 und COP1/SPA Interaktion lokalisiert werden. Durch die gezielte Mutagenese dieses Motivs konnte die Proteinstabilität im Licht, als auch im Dunkeln untersucht werden.
