AbstractMaintenance of genome integrity is of pivotal importance for the propagation of species. Inorder to faithfully propagate the genomic information, eukaryotes have developed a sophisti-cated signaling network, the DNA damage response (DDR), which converges on the down-stream effector p53 protein for mediating the cell-cycle arrest, enhancing the DNA repair, orinducing apoptosis. In order to identify novel players involved in the p53-mediated DDR, weset up a forward genetic screen using Caenorhabditis elegans as an animal model, in whichthe DDR and the p53 protein is evolutionarily conserved. We found that loss of the eukaryotictranslation initiation factor 4E (eIF4E) homolog IFE-4 (Initiation Factor 4E (eIF4E) family) canreverse the ultraviolet (UV)-induced primordial germ cell (PGC) mitotic arrest and germline de-velopmental defect in the global genome nucleotide excision repair (GG-NER)-compromisedxpc-1 mutant animals, resembling the loss of p53 homolog CEP-1 (C. Elegans P-53-like pro-tein). Although expressed in somatic tissues, IFE-4 acts upstream to regulate the expressionand activity of CEP-1/p53 in the germ cells upon UV irradiation, possibly through its transla-tional targets. Moreover, we provide evidence that fibroblast growth factor (FGF) signaling isimplicated in the communication between germ cells and the somatic gonad for the inductionof somatic IFE-4 expression in response to UV and the subsequent activation of CEP-1/p53 sig-naling in germ cells. Collectively, the current study reveals a cell non-autonomous regulatorymechanism through which the translation initiation in the somatic gonad mediates the DDR ingerm cells.ZusammenfassungDie Instandhaltung der Genomintegrita¨t ist ist von ho¨chster Bedeutung fu¨r die Fortpflanzungvon Spezien. Eukaryoten verfu¨gen u¨ber ein komplexes Signalnetzwerk, bekannt als DNA-Reparaturantwort, das am Effektorprotein p53 zusammenla¨uft. p53 ist ein Transkriptionsfak-tor, der einen Zellzyklus-Arrest einleitet und die DNA-Reparaturkapazita¨t erho¨ht, jedoch imFalle irreparabler DNA-Scha¨den auch Apoptose initiieren kann. In der vorliegenden Studienutzten wir den Fadenwurm Caenorhabditis elegans, einen Modellorganismus, in dem dieDNA-Reparatur-antwort einschließlich des p53 Proteins evolutiona¨r konserviert ist, um in einemgenetischen Screen neue Faktoren in der p53-abha¨ngigen DNA-Reparaturantwort zu identi-fizieren. Wir fanden heraus, dass der Verlust des eukaryotischen Translationsinitiationsfaktor4E (eIF4E)-Homologs IFE-4 (Initiationsfaktor 4E (eIF4E) Familie) den UV-strahlungsinduziertenmitotischen Arrest primordialer Keimzellen (PGCs) und einen Keimbahnentwicklungsdefekt innukleotidexzisionsreparaturkompromittierten xpc-1Mutanten verursacht und damit pha¨notypischeinem Verlust des C. elegans p53-Homologs CEP-1 (C. elegans p53-like protein) a¨hnelt. Ob-wohl IFE-4 in somatischen Geweben exprimiert wird, agiert es oberhalb von CEP-1/p53, umdessen Expression und Aktivita¨t in Keimzellen nach UV-Bestrahlung zu regulieren. Dies geschiehtwahrscheinlich u¨ber translationale Targets von IFE-4. Zudem liefern wir Beweise, dass die Fi-broblast Growth Factor (FGF) -Signalisierung wichtig fu¨r die Induktion der somatischen IFE-4-Expression nach UV-Strahlung und der anschließenden Aktivierung des CEP-1/p53-Signalwegsin Keimzellen ist und daher in der Kommunikation zwischen Keimzellen und der somatischenKeimbahnnische wirkt. Zusammengefasst offenbart unsere Studie einen nicht-zellautonomenRegulationsmechanismus, durch den die Translationsinitiation in der somatischen Keimbahn-nische die DNA-Reparaturantwort in Keimzellen steuert.
