本研究采用组织学和免疫组织化学方法探讨炔雌醚暴露对幼年小鼠睾丸生精细胞增殖与凋亡相关蛋白增殖细胞核抗原(PCNA)、半胱氨酸蛋白酶-3(Caspase-3)表达的影响。将20只4周龄雄性昆明小鼠随机分为4组,适应饲养1周后分别腹腔注射50μL含0、1、10、100mg/kg体重炔雌醚的橄榄油,每天1次,连续1周。小鼠8周龄时,取睾丸称重并制备组织切片,HE染色观察病理学变化,免疫组织化学法测定睾丸组织PCNA、Caspase-3的表达。结果显示,曲精小管的管壁变薄,生精细胞排列紊乱,细胞分层减少,间隙增大,管腔内有大量异常脱落的精母细胞和圆形精子细胞,少见长形精子细胞和精子。与对照组相比,PCNA表达随着炔雌醚剂量的增加而极显著减少(P<0.01),Caspase-3的表达随着炔雌醚剂量的增加而极显著增加(P<0.01)。结果表明,炔雌醚通过调节相关蛋白的表达,抑制生精细胞增殖,促进生精细胞凋亡,从而造成雄性生殖功能损伤
