Identification of related species based on pollen morphology is difficult and is limited primarily to genus or family. Because many pollen grains in glaciers contain protoplasm, genetic information of pollen grains should enable identification of plant taxa below the genus level. Our previous presentation in this symposium showed pollen grains from a glacier could be identified at the subsection level by multiplex PCR. However, the obtained sequence data were limited and the identification at the species level was hence unachieved. Consequently, this study attempted to identify the pollen grains at the species level by whole genome amplification method that may provide sufficient sequence data for the identification. Pollen grains used in this study were Pinus extracted from surface snow (1.8-1.9 m depth) collected from the Belukha glacier in the Altai Mountains of Russia in the summer of 2003. Firast, Whole Genome Amplification (WGA) was performed using a single pollen grain. Then, some regions of the chloroplast genome in the WGA DNA were amplified by polymerase chain reaction (PCR), and DNA products were sequenced to identify the pollen grain. The success rate in obtaining the sequence data in this study was 21.4% (n=28), and this result exceeded the success rate 7.6% of our previous study (Nakazawa et al., 2013) using the PCR method. Pinus is a taxon with approximately 111 recognized species in two subgenera, four sections and 11 subsections. Trees of Pinus sibirica and P. sylvestris are currently found at the periphery of the glacier. The pollen grains from Belukha glacier were identified as subsection Strobus including P. sibirica and section Pinus including P. sylvestris. Moreover, the pollen grain in the subsect. Strobus was finally identified as P. Sylvestris. Also, eight species including P. Sylvestris could be shortlisted for another pollen grains in the subsect. Strobus. The results suggested that the WGA method is useful for the species identification of pollen grains preserved in glaciers.  氷河から見つかる花粉は、他の堆積物試料から見つかる花粉と異なり、細胞内物質（原形質）を残存しているものが多い。このことは、氷河中の花粉から遺伝情報が取得できる可能性を示唆する。従来の花粉分析は、花粉の形態によって分類群を同定するため、形態の類似した近縁種の識別は難しく、科あるいは属レベルでの同定に留まる場合が多かった。氷河中の花粉を DNA 分析しその遺伝情報が得られれば、属より下位の階級で同定が可能となる。著者らは昨年の本シンポジウムで、氷河中のマツ属花粉 1 粒ずつを用いてマルチプレックス PCR をおこない、従来の形態観察では不可能であった亜節レベルでの同定が可能になることを示した。しかしながら得られた遺伝情報は限られており、種の同定までは至らなかった。そこで本研究では、大量の遺伝情報取得が可能となる全ゲノム増幅法を用いて、花粉種の同定を試みた。花粉試料は、ロシア・アルタイ山脈にあるベルーハ氷河にて、2003 年夏に氷河表層（1.8-1.9 m 深）に含まれていたマツ属花粉を用いた。花粉 1 粒ずつの全ゲノム増幅をおこない、その後増幅産物をテンプレートに使用し、葉緑体 DNA をターゲットにした PCR を実施した。そして得られた塩基配列をもとに同定をおこなった。分析の結果、塩基配列の取得成功率は 21.4%（n=28）であり、PCR 法による先行研究のそれを上回った（Nakazawa et al., 2013）。マツ属花粉の下位の階級には、2亜属、4節、17亜節、約 111 種が存在する。そのうち、氷河周辺にはシベリアマツとヨーロッパアカマツが分布する。本研究で用いた花粉は、シベリアマツと同じ Strobus 亜節と、ヨーロッパアカマツと同じ Pinus 節に同定された。さらに、Strobus亜節の花粉のうち 1 粒については、シベリアマツと同定された。また、別の 1 粒については、シベリアマツを含む 8 種まで候補を絞ることが出来た。本研究により、全ゲノム増幅法は、氷河中の花粉の種同定に有効であることが示された。 References Nakazawa, F., J.
