Abstract
目的岩藻多糖多存在于褐藻以及海洋无脊椎动物中,是一种独特的结合有硫酸基团的水溶性多糖,由硫酸基岩藻糖、半乳糖、葡萄糖、木糖、鼠李糖、糖醛酸等单糖组成。研究发现岩藻多糖具有抗癌、增强免疫、抗炎、抑制神经细胞损伤、抗病毒等多种生物活性。与多糖相比,寡糖因水溶性好、易被机体吸收利用等诸多独特的功能,近年来更受人关注,但岩藻寡糖的生物学活性讫今尚没有相关报道。本研究旨在建立岩藻寡糖的制备方法,同时对其功能活性进行初步评价。方法采用毛细管电泳法测定岩藻多糖的单糖组成;用酸水解方法制备岩藻寡糖样品;对岩藻寡糖进行抗氧化活性评价,包括羟自由基清除实验、DPPH自由基清除实验、超氧自由基清除实验、还原活性实验与NO清除实验。结果通过毛细管电泳法确定岩藻多糖主要由Xyl:Glc:Rha:Fuc:Gal:Man:GlcA:GalA(ManA)组成,其摩尔比为1.10:1.00:0.60:15.41:9.56:4.78:5.13:2.13(1.51);对酸解寡糖样品进行质谱检测发现水解产物主要为2-8糖;体外抗氧活性实验结果表明,岩藻寡糖具有良好的自由基清除能力
