Abstract
目的:粒径大小是微观颗粒最重要的性质之一,本论文通过考察粒径大小对免疫学效应的调节作用,期待能为纳微球佐剂的设计提供新的理念和思路。方法:采用SPG膜乳化技术制备出不同粒径大小的壳聚糖纳微球作为颗粒模型,并选用专职抗原提呈细胞(APCs)巨噬细胞株J774A.1,考察了粒径对免疫学效应的影响。结果:与微米球相比,纳米球可以被APCs快速的摄取,表现出最大的内吞数量及颗粒表面积。此外,颗粒的胞内运输方式具有显著的粒径依赖性,纳米颗粒有助于提高溶酶体加工、酶解抗原的能力,并且上调了利于抗原提呈,T细胞活化的细胞因子分泌水平。结论:具有较小粒径的纳球在作为疫苗佐剂时更具有潜在的优势,值得后续工作更多..
