Enchytraeids are extracted from soilsampleswithwetͲfunnelbasedmethods,withorwithoutheat, in temperateclimate zones.The ISO guideline 23611Ͳ3 describes a simple cold wet extraction for enchytraeids. OurexperiencewiththismethodinasubtropicalzoneinBrazilhasshownfrequenthighmortalityduring the extraction process, which is critical for the determination of abundance anddiversity.Therefore,wetestedhotandcoldextractionmethodswithsoilsamplescollectedina fragment of Mixed Araucaria Forest in Colombo City, Brazil, in two occasions betweenSeptemberandNovember,2011.Wecomparedtheenchytraeidabundanceestimatedbycoldandhot extractionbasedon ISO 23611Ͳ3 guidelineusing soil coresof 7 cmofdepth x 5.1diameter. The period for cold extraction was 67 hours in a room cooled to 17°C and hotextractionwas performed for 3 hours in a hot enchytraeid extraction devicewith adaptedfunnels and lamps.Theextractedwormswere immediately counted and thepercentageofdamagedwormswasalsorecorded.HealthywormswerekeptinaPetridishwithathinlayerofsedimentandwaterfromtheextractionprocess inacooledroomand identifiedtogenuslevelonthefollowing3days.Thefirstsamplingresulted, inaverage,ca.12,000enchytraeidspersquaremeter (sqm)extractedbyhotorcoldmethod,coefficientsofvariation (CV)of67and105%respectively.Thesecondsamplingresultedinstatisticallymoreenchytraeidsbyhot(11,565sqm)thancold(6,119sqm)method,CVof44%and60%.Coldmethodshowed11%ofdamaged worms after extraction process in the two occasions. Furthermore, most of thewormsweredeadonthefollowingdayandidentificationwasrestrictedtoonly6%ofthecoldextracted worms. In contrast, the viability of the worms extracted by hot method wasgenerallygoodeven3days later.ThegeneraAchaeta,Enchytraeus,Fridericia,GuaranidrilusandHemienchytraeuswerefound.Atleasttwomoresamplingswillbeperformedinthesamearea to confirm the advantages anddisadvantagesofhot and coldmethodsofenchytraeidextraction.
