Althougn alarge amount of information about storage proteins in cereais like rnaize andwheat is available. many species bave not been characterized in this regard.Better knowledge about storage polypcptides and Iheir gene families in olhe rgrass species can lead to lhe identificanon of gene scquences lhat can be usedfor manipulation of grain quality ÍD economically importam cereal species,We repon results of lhe first phasc of a gene prospection study directed 10idenuficauon of potentially importam endosperm-spccific storage proteingene and promoters through comparative analysis of seed proteins of rnaize.sorghum. pearl millet.jecsinte and tripsacum. We show by SDS-PAGE andimmunological analysis ("westem bloning") wilh policlonal antibodies raisedagamst maize proteins (alfa-, bcta- e gamma-zeins) lhat Ihere are similaritiesand divergences amongst polipctidcs of lhe severa! spccies analyzed. Also. lhewestern blots indicated marked differences in amount of similar polipeptidesof lhe severa! species. These results will help direct our work onidemification, cloning and charaaerization of genes and regulatory sequencespotentially useful in lhe improvemcnt of cereal grain quality through geneticengineering.
