Glutamate dehydrogenase was immobilised onto nylon tube and the activity of the enzyme tube investigated. Two different spacer molecules and two coupling techniques were utilised for immobilising the enzyme. Relatively low activity was retained on the tube, but when the enzyme tube was incorporated into a continuous flow system, and used to assay standard ammonia solution, there was adequate response for measurement. The study indicated the potential use ofimmobilised glutamate dehydrogenase in routine analytical

The advantages of using enzymes inclinical analysis have been discussed (Bergmeyer,1965). The tube was washed withdistilled wa te r.Coupling of GDHBefore coupling, the nylon-spacer tube wasreactivated. Two procedures were adopted. Thenylon-spacer tube was perfused with 5% (w/v)glutaraldehyde in 0.2 M borate pH 8.5 for 10 min,or with 4% (w/v) dimethyl suberimidate in N-ethyl-morpholine for 10 min (Morris et ai, 1975).GDH (bovine from Boehringer ManheimGermany) was dissolved in 0.1 M phosphate bufferpH 7.5. The activated nylon tube was filled withthe enzyme solution and incubated for 3h at 4°.The tube was washed with the coupling buffer andbuffer containing 0.2 M NaC!.MATERIALS AND METHODSGlutamate dehydrogenase (GDH) has beenimmobilised onto collagen membrane, but wasused mainly for kinetic studies (Julliard et ai,1971). Preliminary studies on the immobilisationof GDH in a tubular reactor as a possible replace-ment for the soluble enzyme in clinical analysiswere carried out and the results are reported inthis paper.In the recirculation method, the concentra-tion of reactants was similar to that in the solubleassay. Fig. 2 shows the calibration curvesobtained when aqueous ammonia standards wereassayed using 1 m- and 2 m- tube length. Fig. 3shows the response when tubes prepared by usingdifferent spacer molecules and coupling techniqueswere incorporated into the flow system. Adipicdihydrazide as spacer molecules seemed to pro-duce higher activity tubes. Irnidate coupling alsoresulted in better activity retention, as also shownin Table 1.The pH profIle of the immobilised and solubleenzyme is shown in Fig. 4. The loss inactivity was not unexpected. The enzyme comprises subunits which may be dissociated into in-active subunits (Frieden, 1959). The immobilisa-tion procedure may contribute towards this dis-sociati.on. Covalent attachment usually produceslow activity retention. Glutaraldehyde randomlyre.acts with free amino groups and thus may reactwIth certain active site amino groups or aminogroups essential for comforrnational structure.Irnidate, though less efficient in protein coupling,actually retained higher enzymes activity.The hydrophobicity of the support materialmay further enhance the enzyme denaturation,A. To a largeextent, the right spacer has to be discovered byexperimentation.The applicability of immobilised GDH for usein continuous analysis was investigated by itsincorporation into a continuous flow system. Thel.:a1ibration plots show that the response for therange of ammonia concentration used seemed tocurve when a longer tube was used. This effectwas probably due to the carry-over or sampleinteraction effect of the flow system, accentuatedby exposure of the substrate to the long enzymetube. It can be rectified by using a shorter enzymetube (and thus the necessity of producing higheractivity immobilised enzyme). The range ofammonia concentration used corresponded to thatof urea in blood, and linking of urease and GDH inthe immobilised form for urea analysis should bethe ultimate objective.Fig. 4. pH profiles of soluble and immobilisedGDH.
