The mode offormation oj the products is discussed.INTRODUCTIONcyclising under various conditions, several Nbenzoyl derivatives of erythro 2-amino-1-phenylpropan-1-o1 (la-d) in which the electronicporperties of the benzoyl groups are altered byincorporation of either methoxy or nitrogroups.The products from cyclisations oferythrol-aryll-2-benzamidoalkan-l-ols e.g. (1) in the presenceof phosphorus pentaoxide in inert solventsdepend primarily on the reaction temperature(Fitton etal. 1973). At the temperature ofrefluxingtoluene or xylene, the products are usually translcis mixtures of5-phenyl-4,5-dihydro-oxazoles e.g.(2+3), but at higher temperatures. (e.g. that ofrefluxing decalin), the products are mainlyisoquinolines (4), (sometimes rearranged),(Ardabilchi et al. 1977), and it is well-establishedthat the pathway to the isoquinolines normallyinvolves 4,5-dihydro-oxazole intermediacy (Fittonet al. 1974). On the other hand, the action ofpolyphosphoric acid (PPA) at 100°C leads only totranslcis 4,5-dihydro-oxazole mixtures, usually inhigh yield. Infraredspectra (IR) were measured either as nujol mullsor liquid films with a Perkin-Elmer model 257recording spectrophotometer. IH NMR spectrawere recorded with a Varian A90 spectrometerusing tetramethylsilane as the in ternal standard.Mass spectra (MS) were obtained on an AEIMS12 spectrometer.Preparation oferythro-2-benzamido-l-phenylpropan-l-ols (1, a -d)Erythro-2-benzamido-l-phenylpropan-l-ols (l,a -d) (Table 1) were prepared by acylation of thecorresponding amines as described by Fitton andSmalley (1968).GeneralProcedurefor Cyclisation oferythro-2-benzamido-I-phenylpropan-l-ols (1,a -d);To a suspension of the amide 1 (10 g) in theappropriate solvent (30ml) (Table 2) was addedphosphorus pentaoxide (10 g) and the mixturewas heated under reflux for 1 or 3 h. Aftercooling, crushed ice was added portionwise todestroy excess dehydrating agent and separatethe organic layer. The aqueous layer was washedwith ether (100 ml), then basified with 30%aqueous sodium hydroxide until pH 8. The basiclayer was extracted with ether (4xl00ml) and thecombined extract was washed with water anddried. Excess dehydrating agentwas destroyed by addition of ice-water. Theaqueous layer was basified with 30% aqueoussodium hydroxide until pH 8 and the basic layerwas extracted with ether (4xl00 ml). Thecombined extract was washed with water anddried. Evaporation of the ether gave the mixtureof trans- and cis-4,5-dihydro-oxazole (2 and 3)(Table 2).1-(p-Nitrophenyl)-3-methylisoquinoline (4c).(See Table 3).1-(3,5-Dinitrophenyl)-3-methylisoquinoline (4d).(See Table 3).Trans-m.2-(p-methoxyphenyl)-5-phenyl-4-methyl-4,5-dihydro-oxazole (2d and 3d). (Ardabilchi etal.1979).Trans-cis-2-(3,4-Dimethoxyphenyl)-5-phenyl-4-methyl-4,5-dihydro-oxazole (2e and 3e).(Ardabilchi et at. 1979).RESULTS AND DISCUSSIONThe benzamides (la-d) (Table 1) were synthesisedby benzoylation in aqueous sodium hydroxideand ethers of erythro 2-amino-l-phenylpropan-l-01, itself prepared by the method of Fitton andSmalley (1968) .The benzamide cydisations werecarried out under various conditions, and theresults are given in Table' 2.In the presence of phosphorus pentaoxidein refluxing toluene or xylene for 3 h, thePERTANlKA VOL. 13, NO.3, 1990 391ATAN MOHD. When thesecyclisations were repeated in refluxing decalinonly tars resulted, although low yields of thecorresponding 4,5-dihydro-oxazoleswere isolablewhen the heating period was reduced to 1 h.Increased yields of the 4,5-dihydro-oxazoleswereobtained in both cases when the cyclisations werecarried out in PPA at lOa· for 3 h.However, attempted cyclisations in PPA ofthe nitrobenzamides (lc) and (ld) led only torecovery of the starting materials. Similarly, noreactions occurred when the cyclisations werecarried out using phosphorus pentaoxide inrefluxing toluene for 3 h. However, in refluxingxylene, both amides (lc) and (ld) gaverespectively isoquinolines (4c) and (4d) in yieldsof 17% and 16% which were increased to 28%and 35% respectively when refluxing xylene wasreplaced by refluxing decalin for a similar 3 hheating period.Attempted transformations of the 4,5-dihydro-oxazole mixtures (2a + 3a) and (2b +3b)into the corresponding isoquinolines, byheatingwith phosphorus pentaoxide in refluxingdecalin were unsuccessful with only intractabletars being obtained.The products were identified from their IHn.m.r. spectra. The trans/ciY4,5-dihydro-oxazoleisomer mixture were not separated, but the isomerratios were established from the integration ofthe 4,5-dihydro-oxazole 5-methyl proton signals(doublets), those of the cis-isomers appearingupfield (/) - 0.9 p.p.m.) compared with those ofthe corresponding protons of the trans isomerswhich appear at 8 - 1.5 p.p.m.Mechanistic ImplicationsSince the products from the various cyclisationswere exclusively 5-phenyl-4,5-dihydro-oxazolemixturesfrommethoxybenzamides (la) and (lb)or isoquinolines from nitrobenzamides (lc) and(1d), the course of the reaction clearly dependson whether the aryl ring A (see Scheme) is overallelectron-donationgor electron-attracting. On thebasis of 4,5-dihydro-oxazole intermediacy, theformation of isoquinolines from l-aryl-2-benzamidoalkan-l-ols involves three distinctstages and the opposing effects of the methoxyand nitro groups on ring A are likely to beimportant in each stage.5-Phenyl-4,5-dihydro-oxazole formation. - Twomechanisms ('a' and 'b' in Scheme) have beenproposed (Welsh 1949) to explain the formationof trans and ciY4,5-dihydro-oxazoles from erythroamides, and the general predominance of thetrans isomer in the products from these reactions,indicates that mechanism 'a' operatesoverwhelmingly. If A is electron-donating, thismechanism would clearly be favoured and trans-4,5-dihydro-oxazole formation would bepromoted. The opposite would be true when A iselectron-withdrawing.5-Phenyl-4,5-dihydro-oxazole ring-opening - Thisprocess leads to a vinylamide (e.g. 5) and willclearly be favoured when A is electronwithdrawing. (~'l!D Stage 2 1~(3) 01Me(4) • 0---..-/Stage 3 l8A I~(6)Isoquinoline formation - Cyclisation of thevinylamide (6) resulting from stage 2, to anisoquinoline will be favoured when A is electron-withdrawing, but hindered when it is electron-donating.The results can thus be rationalised on theabove basis. Cyclisations involving erythro amides(Ia) and (Ib) lead one to 5-phenyl-4,5-dihydro-oxazoles (mainly trans) since the reactionsproceed via mechanism 'a', Ring-opening of theresulting 4,5-dihydro-oxazoles and eventualcyclisation of the vinylamides are bothdisfavoured.On the other hand, the alternative but less-common pathway Cb') to the 4,5-dihydro-oxazolewill be favoured for erythro amides (I c) and (I d)but the resulting (presumably cis) 4,5-dihydro-oxazoles are not observed since they are likely toundergo rapid ring-opening, with subsequentcyclisation of the resulting vinylamides toisoquinolines also being favoured.ACKNOWLEDGEMENTSWe would like to thank Universiti PertanianMalaysia for financial support, (to A.M.S), andPuan Noraini and Puan Hapsah for typing themanuscript.REFERENCESARDABILCHI, N., A.O., FInoN, j.R.
