1. Zusammenfassung  Fibrilline (-1 und-2) sind 350 kDa große, ubiquitär exprimierte, extrazelluläre Glykoproteine, die in Geweben zu Fibrillin Mikrofibrillen (FMF) assemblieren. FMF gehören zum Hauptbestandteil von elastischen Fasern, kommen aber auch als elastinfreie Fasern, z.B. in der Haut als sogenannte Oxytalanfasern in der dermo-epidermalen Kontaktzone der papillären Dermis vor. FBN1 Mutationen führen zu erblichen Erkrankungen mit überlappenden und gegensätzlichen Ausprägungen, welche als „Fibrillinopathien“ bezeichnet werden. Die Ausprägungen reichen dabei von dünner bis dicker und hyperelastischer bzw. fragiler bis harter Haut. Dabei ist unklar, wie Fibrilline die hierbei zugrundeliegenden Wachstums- und Differenzierungsprozesse steuern. Die plausibelste Erklärung ist, dass Fibrilline neben der strukturellen Integrität von Geweben auch die Aktivität von Wachstumsfaktoren regulieren, welche essentiell fϋr die Homöostase in der Haut sind. Vorherige Studien konnten bereits biochemisch zeigen, dass FMF Wachstumsfaktoren der „transforming growth factor-β“ (TGF- β) Superfamilie in der extrazellulären Matrix (EZM) binden können. Ziel dieser Arbeit war es zu untersuchen, in welchem Maße strukturelle Beeinträchtigungen der FMF in der Haut sich auf die Bioaktivität von Wachstumsfaktoren auswirken. Dabei wurden die Effekte auf die Hauthomöostase mit Hilfe von zwei Fbn1-defizienten Mausmodellen, einer „Knock-in“ Mutante, „green from founder mouse 8“ (GT8), welche nur die N-terminale Hälfe von Fbn1 exprimiert und einer Fbn1+/- Haploinsuffizienz-Mutante, „marfan gene null“ (mgN) untersucht. Die GT8 Haut zeigte eine progressive postnatale FMF Fragmentierung, was zu einer Dysregulierung der BMP Aktivität führte. Mit Hilfe von neu entwickeltem Bioaktivitätsassay konnte in in vitro Zellkulturen bestätigt werden, dass GT8 FMF nicht die nötige strukturelle Integrität haben, um „bone morphogenetic proteins“ (BMP) erfolgreich zu sequestrieren. Ferner konnte mittels Immunfluoreszenz- und Elektronenmikroskopie gezeigt werden, dass eine strukturelle Störung der FMF zu einer veränderten Integrität der elastischen Fasern und Lokalisation ihrer assoziierten Proteine führte. Zudem hatte die Trunkierung der Fibrillin-1 Proteine ultrastrukturell eine Beeinträchtigung der Basalmembranintegrität in Form von lokalen Aufspaltungen, sowie veränderte Hemidesmosomenmorphologie zur Folge. Wider Erwarten zeigte die bisher als phänotypfrei geltende Fbn1+/- Maus im Alter von einem Jahr einen fibrotischen Phänotyp in der Haut mit erhöhtem Kollagengehalt und daraus resultierender erhöhter Steifigkeit, verstärkter lokaler TGF-β Aktivität und epidermaler Hyperproliferation. Im Gegensatz dazu führte die GT8 Mutation zur pathologisch veränderten Quervernetzung von Kollagenen, was mit einer Expressionsänderung von Lysylhydroxylaseenzymen einherging. Auch waren die elastischen Fasern fragmentiert, was zu hyperelastischer Haut führte.  Diese Ergebnisse lassen darauf schließen, dass die strukturelle Integrität der FMF den Aktivierungsstatus von Wachstumsfaktoren reguliert und so unterschiedliche Mutationen zu unterschiedlichen Pathogenesen mit gegenteiligen Ausprägungen führen können. Abstract Fibrillins (-1 and-2) are 350 kDa large, ubiquitously expressed extracellular proteins which assemble into fibrillin microfibrils (FMF) in tissues. FMF are major building blocks of elastic fibers, but can also be found in fibers devoid of elastin, e.g. in oxytalan fibers of the dermal-epidermal junctions of the skin. FBN1 mutations result in congenital genetic disorders with common and opposing phenotypes summarized as “fibrillinopathies“. The phenotypes range from thin to thick and from hyperelastic and fragile to stiff skin. Yet, it is unclear how fibrillins regulate these various growth and differentiation processes. The most plausible explanation could be that FMFs not only influence the structural integrity of tissues but also the activity of growth factors that are essential for skin homeostasis. Recent studies showed that FMF are able to sequester the growth factors of the transforming growth factor-β (TGF-β) super family in the extracellular matrix (ECM). The aim of this study was to analyze to what extent the structural impairment of FMF in the skin has an impact on the bioactivity of growth factors. For this purpose a knock-in mutant mouse model that carries the N-terminal half of Fibrillin-1, green from founder mouse 8 (GT8), and a Fbn1+/- haploinsufficient mouse model, marfan gene Null (mgN) were analyzed. GT8 skin showed a progressive postnatal FMF fragmentation resulting in a dysregulation of BMP activity. Using a newly established bioactivity assay it could be shown in vitro that due to structural impairment GT8 FMF are not able to sequester bone morphogenetic proteins (BMP). Moreover, immunofluorescence and electron microscopy analyses showed that structural impairment of FMF led to altered integrity of elastic fibers and associated proteins. In addition, truncation of Fibrillin-1 proteins resulted in ultrastructural impairment of basement membrane integrity with local microblistering and altered hemidesmosomal morphology. Unexpectedly, the Fbn1+/- mouse, that has been previously described to lack a phenotype, showed a fibrotic phenotype in skin accompanied by elevated collagen content resulting in increased skin stiffness, locally increased TGF-β activity and epidermal hyperproliferation. In contrast, the GT8 mutation led to pathologically altered collagen cross-linking associated with elevated lysyl hydroxylase expression. In addition, the fragmentation of elastic fibers led to hyperelastic skin. These results suggest that the structural integrity of FMF regulates the activation state of growth factors. Thus, different mutations result in different pathogeneses with distinct phenotypes.
