本計畫之目的在探討大豆種子發芽期間耐淹水 逆境的機制及遺傳.試驗以種子具耐淹水能力差 異的20個品系為材料,首先將調查種子物理特性, 包括各品系種子大小,種子吸水速率,種皮百分率 等性狀與耐淹水性之關係,並應用電子顯微鏡來 觀察種子於淹水前,淹水一天及淹水三天後在構 造上的變化,同時進行耐與不耐淹水的品系間之 雜交十個組合,供為耐淹水機制之遺傳解析.第二 年以電泳進行生化特性分析,以了解Alcohol Dehydrogenase(ADH),Pyruvate decarboxylase(PDC),Lactatedehydrogenase(LDH)及Malate dehydrogenase(MDH)等酵素活性 在淹水一天及淹水三天後之差異,並進行單向或 雙向電泳解析各淹水前後之總蛋白質,試圖尋出 與耐淹水特性有關的生理,生化機制.第三年以雜 交所得F1及F2的種子為材料進行與耐淹水能力有密切關係的物理性狀及生化性狀的遺傳解析
