Tissue specific functions of Caenorhabditis elegans DAF-16/FOXO in longevity and stress response

Abstract

进化保守的胰岛素信号通路是调控秀丽隐杆线虫发育、衰老和压力应激的重要信号通路之一。胰岛素受体DAF-2位于胰岛素信号通路的上游,它可与胰岛素结合来感受外界信号刺激,daf-2突变显著延长线虫寿命并增加其抗压性;FOXO转录因子DAF-16位于胰岛素信号通路的最下游,daf-2突变和压力条件促使它进入细胞核,进而调控靶基因表达,而daf-16突变可完全抑制daf-2突变所引起的寿命延长和抗压性。已有研究暗示功能重要的daf-16亚型daf-16b/c和daf-16d/f在不同组织中表达,并可能在功能上存在差异。为进一步确定daf-16在不同组织中的功能,本文首先构建了daf-2突变体背景上组织特...Evolutionary conserved Insulin/IGF-1 signaling(IIS) pathway is one of the most important pathways that regulate development, aging and stress response in Caenorhabditis elegans.
