Reversal of multidrug resistance in human hepatocellular carcinoma cells by JNK

Abstract

目的:多药耐药(multidrugresistance,MDR)细胞的存在是肝癌化疗失败的重要原因。有研究表明抗肿瘤药物能引起JNK信号传导系统的变化,提示通过调节JNK有可能逆转MDR。本课题应用抗癌药物阿霉素(ADM)诱导建立人肝癌多药耐药细胞HepG2/ADM,通过增高和降低肝癌多药耐药细胞HepG2/ADM中JNK的表达,检测多药耐药细胞的耐药性的变化,初步探讨JNK是否可做为逆转肝癌多药耐药细胞靶点的依据。 方法:采用药物浓度梯度递增诱导法诱导亲本细胞建立人肝癌多药耐药细胞系HepG2/ADM,流式细胞仪分析细胞周期并检测多药耐药相关蛋白P-gp(P-glycoprotein)和M...Objective: Multidrug resistance (MDR) is almost constantly expressed in hepatocellular carcinoma(HCC) and represent one of the major problems for cancer eradication by limiting the efficacy of chemotherapy.
