Investigation on the toxicity interaction mechanisms of Benzalkonium Chloride on the Rabbits’ cornea

Abstract

目的:观察苯扎氯氨对兔角膜急性毒性作用并探讨其机制。方法:按照局部点眼的方法,分别给兔滴用不同浓度的苯扎氯氨(0.01%、0.05%和0.10%),每天两次,连续4天。对侧未治疗眼做为对照。裂隙灯显微镜及泪液分泌实验(Scherimer’s)观察其眼表及泪膜变化。共焦显微镜观察活体角膜各层细胞结构变化。观察角膜MUC5AC、ZO-1表达及分布情况,结合荧光渗透试验(CF)了解其对角膜内皮屏障结构及功能的影响。结果:裂隙灯显微镜及Scherimer’s实验未发现泪膜及角、结膜改变;角膜切片HE检查见角膜各层细胞排列整齐,未见明显炎症改变;共焦显微镜检查发现角膜上皮细胞层数增多、表层细胞横径缩小、...Object: To invest the acute toxicity effect and analyze the mechanism of Benzalkonium Chloride on rabbits’ cornea. Methods: We have used the topical application of kinds of concentrations (0.01%.0.05%.0.10%)of Benzalkouium Chloride as the obversation and the contralteral as control .
