Isolation of microsatellite markers, construction of genetic linkage maps and QTL mapping of growth-related traits in small abalone, Haliotis diversicolor

Abstract

本研究采用磁珠富集法,用14个探针构建了杂色鲍微卫星富集文库。从文库中随机选取768个阳性克隆进行测序,拼接得到729个重叠群,其中656个重叠群包含微卫星序列。利用微卫星两侧保守区域设计了583对微卫星引物,从中筛选出退火温度在50-64℃,PCR产物稳定扩增、条带清晰的微卫星标记308个。随机选取其中的11个微卫星标记用于标记质量评估,这11个微卫星标记的PIC值在0.7276到0.9163之间,具有高度多态性。 利用开发的308个微卫星标记,以养殖个体♀和野生个体♂杂交F1家系为作图群体,构建了杂色鲍的遗传连锁图谱。308个微卫星标记中有182个标记在亲本中具有多态性,可以用于图谱构建...Small abalone, Haliotis diversicolor is naturally distributed along the coastal waters of East Asia: from Japan to the Philippines.
