Abstract

P-TEFb是由CDK9及其调节亚基CyclinT1组成的异二聚体激酶,存在于所有的真核生物中。研究证明:P-TEFb是基因转录从起始阶段进入有效延伸阶段的必需因子,与艾滋病、心肌肥大和肿瘤发生均有密切关系。在RNA聚合酶Ⅱ(PolⅡ)介导的mRNA转录过程中,P-TEFb被募集到基因启动子区,通过磷酸化PolⅡ及延伸抑制因子,从而刺激全长mRNA的高效转录。在细胞内,P-TEFb以无活性和有活性(可募集)两种复合物形式存在。本实验室前期系列的研究发现,细胞在受到外界胁迫因素刺激后,可激活细胞内钙离子-PP2B及PP1信号途径,通过PP2B和PP1蛋白磷酸酶的协调去磷酸化作用,导致无活性复合物...P-TEFb, a heterodimer kinase consisting of CDK9 and its regulator CyclinT1, exists in all eukaryotic species. P-TEFb has been demonstrated to be a key player in the transition from transcription initiation to the productive elongation phase during gene transcription.
