Differential Expression of Periostin in Human Breast Carcinomas Promotes Cancer Cells Survival

Abstract

Periostin是一种分泌蛋白,由N末端信号肽、半胱氨酸富含区、四个内部同源结构域和C末端的亲水结构域组成,在骨的形成和损伤修复中起着重要作用。它与昆虫神经系统中粘附分子fasciclinⅠ具有很高的结构同源性。近年来人们还发现periostin在卵巢癌、非小细胞肺癌等肿瘤组织中高表达.特别是在结肠癌中,periostin蛋白的高表达能够促进癌细胞生存从而促进结肠癌的发展和转移。我们用迴文PCRcDNA差异显示的方法发现periostin基因在乳腺癌组织中高表达,并通过Northernblot和免疫组化方法进一步验证了这一结果。随后,我们构建了稳定表达periostin蛋白的人乳腺癌细胞MC...Periostin is a secreted protein containing an N-terminal secretory signal peptide, followed by a cysteine-rich domain, four internal homologous repeats and a C-terminal hydrophilic domain.
