目的:研究CD4+T淋巴细胞上Kv1.3通道在大鼠AS疾病过程中的作用,探讨特异性阻断Kv1.3通道对大鼠AS病情的影响以及可能的机制。 方法:实验动物分为对照组、模型组和药物处理组。模型组给予初始维生素D3负荷量腹腔注射,联合高脂饲料持续喂养(8周);药物处理组在模型组的基础上加用特异性Kv1.3通道阻断剂ShK(L5),10ug/kg/d,皮下注射,持续8周。分离大鼠脾脏淋巴细胞,流式细胞术检测脾脏淋巴细胞中CD4+T细胞在总T淋巴细胞中所占的比例。利用免疫磁珠法分选出脾脏中的CD4+T细胞:流式细胞术检测其中CCR7-的TEM细胞所占比例的变化;提取RNA,实时荧光定量RT-PCR检测...Objective: To investigate the role of voltage-gated potassium channel Kv1.3 of CD4+ T lymphocytes in rat’s atherosclerosis, find out whether the selective Kv1.3 blocker ShK(L5) can alleviate rats’ atherosclerosis, and to discuss how the selective Kv1.3 blocker affect the progress of atherosclerosis.
