Proteomics Approach for Outer Membrane Proteins from Important Gram Negative pathogens

Abstract

采用蛋白质组学、基因工程技术、生物信息分析等研究方法,对10种重要的革兰氏阴性病原菌的外膜蛋白进行了较系统的研究,成功建立了6种细菌的分子表达图谱,重点筛选和鉴定了与盐调节、耐药和交叉抗原相关的蛋白质组。在对外膜蛋白质提纯方法比较的基础上、应用月桂酰肌氨酸钠二步法获得的高纯度外膜蛋白质,建立六种革兰氏阴性菌外膜蛋白的分子解剖图谱。通过对已知蛋白的网上模拟及鉴定、双向电泳图谱的比较,建立了一种对未进行全基因序列测定的细菌进行PMF鉴定的方法,并在溶藻弧菌及荧光假单胞菌这两种菌中得到了成功的应用。通过比较蛋白质组学方法,在国内外首次从副溶血弧菌及溶藻弧菌中发现和鉴定了一种类似于大肠杆菌双成分调控系...With the rapid development of microbial genome sequencing project, microbial proteomics has become a powerful tool to elucidate the life mechanism of microorganisms.
