Construction of Recombinant Plasmid for Yeast Ethanolic Xylose-Fermentation

Abstract

燃料乙醇是公认的最有发展前景的可再生清洁能源之一。木质纤维素是地球上最充分的有机资源,以木质纤维素类生物质生产燃料乙醇不仅能够降低燃料乙醇生产的原料成本,同时在废物处理、环境保护等方面起到一定的作用。木糖发酵是植物纤维原料生物转化制取乙醇商业化生产的基础和关键,但自然界存在的微生物菌株不能满足商业化生产的需要。利用基因工程技术对酵母菌进行改造,以提高它们的木糖发酵能力成为目前研究和开发的重点。 采用oligo和Primerpremier5软件设计引物,从休哈塔假丝酵母中克隆到木糖还原酶基因xyl1,从热带假丝酵母中克隆到木糖醇脱氢酶基因xyl2,从酿酒酵母中扩增到木酮糖激酶基因xks1。对三...Fuel ethanol is regarded as one of the most promising environment friendly fuel in the future.
