Preparation of floate: conjugated chitosan nanoparticles wrapped hydroxycamptothecin and study of anti-tumor activity

Abstract

本文在分析了目前国内外靶向纳米药物制剂的研究现状和发展趋势的基础上,用壳聚糖作为载体、羟基喜树碱作为原料药,采用复合乳液溶剂挥发-界面交联方法制备出了叶酸偶联壳聚糖载羟基喜树碱纳米药物制剂(FA-NPs)。经性能表征、体外释药性能研究、肿瘤细胞摄取实验、体内抗肿瘤活性和组织分布研究,优化制备的FA-NPs平均粒径为250nm、包封率和载药量分别为81.2%和2.524%,体外累计释药时间和抗肿瘤活性均优于现有羟基喜树碱制剂,FA-NPs在体内正常组织分布和全身毒副作用却少于羟基喜树碱制剂。主要研究结果如下: 1.以壳聚糖作为药物载体、叶酸作为靶向配体、羟基喜树碱为药物模型,采用复合乳液溶剂挥...Based on the analysis of research and development trends of the current targeting nano-drug preparation at home and abroad, a folate-conjugated chitosan nanoparticles wrapped hydroxycamptothecin agents (FA-NPs) by chitosan as a carrier, hydroxycamptothecin as a raw material medicine and using the method of composite emulsion solvent evaporation - interface for cross-linking are presented in this t...学位:博士后院系专业:生命科学学院生物医学科学系_生物医学工程学号:BH1700020
