The mechanism of orphan receptor TR3 regulated by methyltransferase PRMT1

Abstract

蛋白精氨酸甲基转移酶1(proteinarginine-methyltransferase1,PRMT1)负责哺乳动物中至少85%蛋白精氨酸底物的甲基化。蛋白甲基化修饰可以调节组蛋白和非组蛋白的各种功能,包括转录活性、蛋白与DNA和RNA之间的相互结合、以及蛋白的亚细胞定位等。TR3是类固醇激素受体超家族成员,迄今为止未发现其特异性配体,所以又称为孤儿受体。TR3作为转录因子从转录和转录后水平上进行调控,从而在细胞增殖与凋亡过程中发挥关键作用。本文研究中,我们发现PRMT1不能甲基化TR3,但是PRMT1促进TR3启动子的转录、提高TR3mRNA的表达水平和延缓TR3蛋白的半衰期,从而提高TR...PRMT1 (protein arginine-methyltransferase 1,PRMT1),a predominant memberof arginine methyltransferases, is responsible for at least 85% of all argininemethylation reactions in mammal cells, and thus plays important roles on cellularprocesses, including transcriptional regulation, interaction between protein with DNA and RNA, and cellular location of protein.
