Construction of Thermotolerant Engineering Yeast Strain and Analysis of Ethanol Production Fermenting with Hydrolysis Liquid of Cellulose

Abstract

随着石油资源日渐匮乏和全球环境污染问题的日益加重,各国纷纷开始开发新型能源。纤维素是地球上最丰富、最廉价的可再生资源。利用生物技术将纤维素转化为酒精,不仅节约资源,而且能够实现环境的保护。 本实验采用酶解法制备了葡萄牙假丝酵母(Candidalusitaniae)和酿酒酵母(Saccharomycescerevisiae)原生质体,并研究了酶浓度、酶解时间、酶解温度、预处理、渗透压稳定剂及pH值等因素对其形成和再生的影响。结果表明,葡萄牙假丝酵母原生质体形成的最佳条件为:菌龄8h,酶浓度3.0U/mL,酶解温度30℃,山梨醇(1.0mol/L,pH6.5)为渗透压稳定剂,酶解时间75min;...Many countries started to explore new energy due to the shortage of fossil fuel resources and the increasing serious environmental pollution.
