本文以五唇兰、蝴蝶兰为材料,应用石蜡切片和花粉压片观察了2种兰花雄配子体的发育,进行了精细胞的分离;在分离生活胚囊的基础上,进行了卵细胞、合子和多细胞胚的分离研究。本文的主要结论是:1.五唇兰、蝴蝶兰的小孢子四分体均有四面体型、左右对称形、交叉形和T形等多种类型,它们发育成四合花粉,并由四合花粉聚集成花粉块。花粉成熟时为二细胞型。2.通过人工授粉使花粉管在子房内发育,再利用花粉管在甘露醇中直接爆破分离出五唇兰、蝴蝶兰的精细胞,成对的2个精细胞在体积大小、荧光强弱均显示有较大差异。应用显微操作仪可收集精细胞群体。3.应用酶解-振荡法处理胚珠,分离出2种兰花的生活胚囊,继之以显微解剖分离出2种兰花...Based on the materials of Doritis pulcherrima and Phalaenopsis hybrid, the development of male gametophytes were observed by the methods of paraffin sections and pollen compression.
